NVP-BEZ235誘導(dǎo)多囊腎大鼠膽管上皮細(xì)胞自噬的機(jī)制研究

【摘要】:目的:探討PI3K/Akt/m TOR雙靶點(diǎn)抑制劑NVP-BEZ235誘導(dǎo)多囊腎(polycystic kidney,PCK)大鼠膽管上皮細(xì)胞自噬的作用。方法:免疫組化法檢測(cè)p-m TOR和p-Akt在PCK大鼠膽管上皮細(xì)胞中的水平。WST-1比色法檢測(cè)NVP-BEZ235對(duì)膽管細(xì)胞活力的抑制作用以及LC3、Beclin 1基因沉默和自噬特異性抑制劑3-甲基腺嘌呤(3-methyladenine,3-MA)對(duì)細(xì)胞活力的影響。蛋白免疫印跡法檢測(cè)NVP-BEZ235對(duì)PI3K/Akt/m TOR信號(hào)通路相關(guān)蛋白及自噬標(biāo)志蛋白LC3和Beclin 1的變化。結(jié)果:p-m TOR和p-Akt在PCK大鼠膽管上皮細(xì)胞中顯著升高,NVP-BEZ235可明顯抑制膽管上皮細(xì)胞的活力,且呈濃度和時(shí)間依賴(lài)性改變(P0.05);NVP-BEZ235明顯抑制PI3K/Akt/m TOR信號(hào)通路相關(guān)蛋白的水平;并上調(diào)自噬標(biāo)志蛋白LC3 II/LC3 I比值和Beclin 1蛋白的表達(dá)水平。LC3、Beclin 1基因沉默和3-MA均可明顯減弱NVP-BEZ235對(duì)細(xì)胞活力的抑制作用(P0.01)。結(jié)論:NVPBEZ235可抑制PCK大鼠膽管上皮細(xì)胞的活力,其機(jī)制與自噬密切相關(guān)。
