LRPPRC對前列腺癌細胞凋亡的影響

【摘要】:目的:分析富含亮氨酸的三角狀五肽重復結(jié)構(gòu)蛋白(LRPPRC)和抗凋亡蛋白Bcl-2在前列腺癌組織的表達及LRPPRC對前列腺癌細胞凋亡的影響。方法:應用免疫組化法檢測198例前列腺癌組織中LRPPRC和Bcl-2的表達;分析LRPPRC和Bcl-2表達的相關(guān)性;分析LRPPRC和Bcl-2表達與預后的關(guān)系;應用si RNA LRPPRC轉(zhuǎn)染前列腺癌細胞下調(diào)LRPPRC水平,觀察對細胞增殖、凋亡和Bcl-2、Bax、caspase-3蛋白表達的影響。結(jié)果:前列腺癌和前列腺增生組織LRPPRC的表達率分別為61.6%(122/198)和18.0%(18/100)(P0.001);Bcl-2的表達率分別為49.5%(98/198)和45.0%(45/100)(P=0.539)。LRPPRC和Bcl-2的表達有顯著正相關(guān)性(r=0.179,P=0.012)。LRPPRC和Bcl-2同時高表達的患者無疾病進展生存和總生存期均顯著降低(P0.001)。下調(diào)LRPPRC表達可抑制前列腺癌細胞增殖,促進前列腺癌細胞凋亡,降低Bcl-2蛋白的表達,使caspase-3活化增多,對Bax表達無顯著影響。結(jié)論:前列腺癌組織LRPPRC表達顯著增高;LRPPRC與抑制凋亡有關(guān);LRPPRC在前列腺癌中的作用機制與抗凋亡蛋白Bcl-2有關(guān)。
[Abstract]:Aim: to analyze the expression of trigonometric pentapeptide repeat protein (LRPPRC) and anti-apoptosis protein Bcl-2 rich in Leucine in prostate cancer and the effect of LRPPRC on apoptosis of prostate cancer cells. Methods: the expression of LRPPRC and Bcl-2 in 198 cases of prostate cancer was detected by immunohistochemistry, the correlation between the expression of LRPPRC and Bcl-2 was analyzed, and the relationship between the expression of LRPPRC and Bcl-2 and prognosis was analyzed. Si RNA LRPPRC was used to down-regulate the level of LRPPRC in prostate cancer cells, and the effects on cell proliferation, apoptosis and Bcl-2,Bax,caspase-3 protein expression were observed.
