骨髓間充質(zhì)干細(xì)胞經(jīng)腎動脈途徑不同移植次數(shù)對大鼠阿霉素腎病的修復(fù)作用比較
[Abstract]:Aim to compare the effects of bone marrow mesenchymal stem cells (BMSC) on renal function after transplantation of bone marrow mesenchymal stem cells (MSCs) through renal artery in rats with adriamycin nephropathy. Methods Thirty-two rats were randomly divided into three groups: M _ 0, M _ 1, M _ 2, M _ 3 (4 脳 n, n = 8) and control group (n = 8). BMSC cells were transplanted 0 times (M _ 0 group), 1 time (M _ 1 group), 2 times (M _ 2 group) and 3 times (M _ 3 group) with 2 脳 10 ~ 6 cells / ml, for 0.5 ml. No BMSC transplantation group was performed with L-DMEM medium 0.5 ml as placebo control, the interval of transplantation was 1 week. Blood urea nitrogen, serum creatinine, 24-hour urine protein and 24-hour urine microprotein were measured before transplantation and 1 and 2 weeks after the last transplantation. At the end of the first week after the last transplantation, pathological slices were made and the pathological changes and distribution of BMSC cells in the kidney of rats were observed under laser focusing microscope. Results the levels of blood urea nitrogen, serum creatinine, 24 h urinary protein and 24 h urine microprotein in M0, M1, M2 and M3 groups were significantly higher than those in N group at the end of the first week of transplantation (P0.001). At the end of the first week after the last transplantation, the total urine protein content in M3 group and M3 group was significantly higher than that in N group (P0.001). 24 h urinary microprotein was significantly lower than that of M1 group (P0.05), while there was no significant difference between M2 and M3 groups (P < 0. 063). Conclusion Transplantation of BMSC through renal artery has the best effect on adriamycin nephropathy in rats.
