GPC5異常表達(dá)對前列腺癌細(xì)胞生物學(xué)行為的影響

【摘要】:目的 研究GPC5異常表達(dá)對前列腺癌細(xì)胞生物學(xué)行為的影響,并對其相關(guān)分子機(jī)制進(jìn)行初步探討。 方法 1.構(gòu)建GPC5高/低表達(dá)質(zhì)粒,轉(zhuǎn)染前列腺癌細(xì)胞PC3及LNCap,通過與對照組比較,觀察分析GPC5異常表達(dá)對前列腺癌細(xì)胞增殖和侵襲的影響。2.通過提高前列腺癌細(xì)胞PC3中c-Myc的表達(dá),觀察c-Myc過表達(dá)對前列腺癌細(xì)胞PC3中GPC5表達(dá)的影響。3.利用免疫共沉淀技術(shù)及質(zhì)樸分析,尋找前列腺癌細(xì)胞中與GPC5相互作用的生長因子/受體配體。 結(jié)果 1.上調(diào)GPC5表達(dá)可顯著抑制前列腺癌細(xì)胞PC3的增殖和侵襲能力;下調(diào)GPC5使前列腺癌細(xì)胞LNCap增殖及侵襲能力顯著增強(qiáng)。 2.在前列腺癌細(xì)胞PC3中,c-Myc的上調(diào)伴隨著GPC5的下調(diào), 3.免疫共沉淀及蛋白質(zhì)樸分析顯示在前列腺癌細(xì)胞PC3中,肝癌衍生生長因子(HDGF)與GPC5相結(jié)合而發(fā)揮作用。 結(jié)論 1.GPC5異常表達(dá)可影響前列腺癌細(xì)胞PC3及LNCap的細(xì)胞生物學(xué)行為,證實(shí)GPC5在前列腺癌發(fā)生發(fā)展中發(fā)揮重要作用,可能成為治療前列腺癌的一個(gè)潛在的靶點(diǎn)。 2.c-Myc可調(diào)節(jié)前列腺癌中GPC5的表達(dá)量,進(jìn)而影響前列腺癌的發(fā)生發(fā)展。 3.前列腺癌細(xì)胞膜表面的GPC5可能作為外源性HDGF在細(xì)胞膜表面的共受體并介導(dǎo)其進(jìn)入細(xì)胞內(nèi)發(fā)揮作用。
[Abstract]:Objective to investigate the effect of abnormal expression of GPC5 on biological behavior of prostate cancer cells and its molecular mechanism. Method 1. High / low expression plasmids of GPC5 were constructed and transfected into prostate cancer cells PC3 and LNCap,. The effects of abnormal expression of GPC5 on proliferation and invasion of prostate cancer cells were observed and analyzed. By increasing the expression of c-Myc in prostate cancer cell line PC3, the effect of c-Myc overexpression on the expression of GPC5 in prostate cancer cell line PC3 was observed. 3. 3. By means of immunoprecipitation and simple analysis, the growth factor / receptor ligand interacting with GPC5 in prostate cancer cells was found. Result 1. Upregulation of GPC5 expression significantly inhibited the proliferation and invasion of prostate cancer cell line PC3, and down-regulation of GPC5 significantly enhanced the proliferation and invasion of prostate cancer cell line LNCap. 2. In prostate cancer cell line PC3, the up-regulation of c-Myc was accompanied by down-regulation of GPC5. Immunoprecipitation and proteomic analysis showed that hepatoma derived growth factor (HDGF) combined with GPC5 in prostate cancer cell line PC3. Conclusion the abnormal expression of 1.GPC5 may affect the biological behavior of PC3 and LNCap in prostate cancer cells. It is confirmed that GPC5 plays an important role in the development of prostate cancer and may become a potential target for the treatment of prostate cancer. 2.c-Myc can regulate the expression of GPC5 in prostate cancer, and then affect the development of prostate cancer. 3. GPC5 on the surface of prostate cancer cell membrane may act as a co-receptor of exogenous HDGF on the cell membrane and mediate its entry into the cell.
