海昆腎喜膠囊和百令膠囊對(duì)尿酸性腎病的影響

【摘要】:目的:探討海昆腎喜膠囊及百令膠囊對(duì)高尿酸腎病的防治作用及其可能的作用機(jī)制。方法:6周齡SPF級(jí)雄性Wistar大鼠93只,普通飼料適應(yīng)性喂養(yǎng)1周后,隨機(jī)分為正常組、高尿酸血癥組、海昆腎喜干預(yù)組、百令膠囊干預(yù)組。高尿酸組給予腺嘌呤灌胃50mg/(kg.d),高嘌呤飼料喂養(yǎng),同時(shí)給予氧嗪酸鉀乳懸液4ml/(kg. d)分兩次皮下注射;干預(yù)組大鼠在上述飼養(yǎng)給藥的基礎(chǔ)上同時(shí)給予相應(yīng)的藥物灌胃治療;正常組大鼠給予普通飼料飼養(yǎng)作為正常對(duì)照。在實(shí)驗(yàn)的第2、4、6周每組隨機(jī)選取6只大鼠,內(nèi)眥靜脈取血,檢測尿酸(UA)、尿素氮(BUN)、肌酐(CR)及肌酐清除率(Ccr);收集大鼠24h尿液,測定24h尿微量白蛋白定量;同時(shí)每組處死2只大鼠,測定左腎質(zhì)量指數(shù),取腎臟組織行HE, MASSON染色,免疫組化測定白介素1-β(IL-1β)、轉(zhuǎn)化生長因子-β(TGF-β1)、α-平滑肌肌動(dòng)蛋白(α-SMA)的表達(dá)。結(jié)果:與正常組比較,高尿酸血癥組在第2、4、6周UA、BUN, CR,24小時(shí)尿蛋白,左腎質(zhì)量分?jǐn)?shù)升高,Ccr降低,結(jié)果有統(tǒng)計(jì)學(xué)意義(p0.01)。與高尿酸血癥組比較,海昆腎喜干預(yù)組在第2,4,6周CR,Ccr,24小時(shí)尿蛋白有明顯降低(p0.01),第6周BUN,左腎質(zhì)量分?jǐn)?shù)有明顯改善(p0.01);百令膠囊干預(yù)組在第2、4、6周Ccr,24小時(shí)尿蛋白有明顯降低,第2、6周BUN有明顯降低,第4、6周CR,左腎質(zhì)量分?jǐn)?shù)有明顯改善(p0.01)。與正常組相比,高尿酸血癥組在第2、4、6周HE及MASSON染色提示大鼠腎間質(zhì)膠質(zhì)纖維增加,腎間質(zhì)組織表達(dá)TGF-β1,IL-1β,α-SMA明顯增加(p0.01)。與高尿酸血癥組比較,海昆腎喜干預(yù)組在第2、4、6周,腎間質(zhì)沉淀及纖維化減輕,TGF-β1,IL-1β,α-SMA表達(dá)明顯減少(p0.01);百令膠囊干預(yù)組在第4、6周,腎間質(zhì)沉淀及纖維化減輕,TGF-β1,IL-1β,α-SMA表達(dá)明顯減少(p0.01)。 結(jié)論:海昆腎喜膠囊及白令膠囊能改善高尿酸腎病大鼠腎臟功能,減少腎間質(zhì)TGF-β1,IL-1β,α-SMA的表達(dá),減輕腎間質(zhì)纖維化。
[Abstract]:Objective: to investigate the preventive and therapeutic effects of Haikunshenxi capsule and bailing capsule on hyperuric acid nephropathy and its possible mechanism. Methods: 93 SPF grade male Wistar rats aged 6 weeks were randomly divided into normal group, hyperuricemia group, Hishenkunxi intervention group and bailing capsule intervention group. In hyperuricemia group, 50mg/ (kg.d) was given orally, and hyperpurine was fed on diet and 4ml/ (kg.) was administered with potassium oxazinate milk suspension. D) two times of subcutaneous injection; intervention group rats on the basis of the above feeding and administration of the corresponding drug infusion treatment; normal group rats were fed with normal diet as normal control. Six rats in each group were randomly selected at the 2nd week and 4th week of the experiment. The blood was collected from the inner canthus vein. The uric acid (UA), urea nitrogen (BUN), creatinine (CR) and creatinine clearance rate (Ccr);) were collected from 24 hours urine of the rats. At the same time, two rats were killed in each group. The left renal mass index was measured. The renal tissues were stained with HE, MASSON, and interleukin-1 尾 (IL-1 尾) and transforming growth factor- 尾 (TGF- 尾 1) were detected by immunohistochemistry. Expression of 偽-smooth muscle actin (偽-SMA). Results: compared with the normal group, the urine protein, left renal mass fraction and Ccr were significantly increased in hyperuricemia group at the 6th week of the 2nd week (p0.01). Compared with the hyperuricemia group, the urine protein of CR,Ccr,24 in the intervention group was significantly decreased at the 2nd week (p0.01), and the left renal mass fraction of the BUN, was significantly improved at the 6th week (p0.01). In the intervention group, urinary protein decreased significantly at the 2nd week, the BUN decreased at the 6th week, and the left renal mass fraction of CR, improved significantly at the 4th week (p0.01). Compared with normal group, HE and MASSON staining in hyperuricemia group at the 2nd week and 4th week showed that the expression of TGF- 尾 1 and IL-1 尾 and 偽 -SMA in renal interstitial tissue was significantly increased (p0.01). Compared with the hyperuricemia group, the renal interstitial sedimentation and fibrosis were alleviated and the expression of IL-1 尾, 偽-SMA decreased significantly in the Hishenhei intervention group at the 4th week (p0.01). In the intervention group, the renal interstitial sedimentation and fibrosis were alleviated, and the expression of IL-1 尾, 偽-SMA of TGF- 尾 1 was significantly decreased in the intervention group at the 4th week (p0.01). Conclusion: Haikunshenxi capsule and Bering capsule can improve the renal function, reduce the expression of TGF- 尾 1 and IL-1 尾, 偽-SMA and alleviate renal interstitial fibrosis in rats with hyperuricemia nephropathy.
