【摘要】：目的通過慢病毒表達(dá)系統(tǒng),使用shRNA下調(diào)人膀胱癌T24細(xì)胞GSTP1基因的表達(dá),研究其對T24細(xì)胞增殖的影響,并探討其可能的作用機(jī)制。方法通過慢病毒表達(dá)系統(tǒng),建立穩(wěn)定低表達(dá)GSTP1基因的T24細(xì)胞株;進(jìn)行CCK-8實驗以及克隆形成實驗,檢測GSTP1表達(dá)下調(diào)對T24細(xì)胞增殖的影響;對細(xì)胞進(jìn)行Annexin V-FITC/PI染色,使用流式細(xì)胞儀,檢測GSTP1表達(dá)下調(diào)對T24細(xì)胞凋亡的影響。結(jié)果與未處理組、對照組相比,GSTP1干擾組的T24細(xì)胞增殖受到明顯抑制,差異有統(tǒng)計學(xué)意義(P0.01);細(xì)胞凋亡檢測結(jié)果顯示,GSTP1表達(dá)下調(diào)后明顯促進(jìn)T24細(xì)胞凋亡,差異有統(tǒng)計學(xué)意義(P0.01)。結(jié)論 GSTP1表達(dá)下調(diào)抑制膀胱癌細(xì)胞T24增殖,并其機(jī)制可能與促進(jìn)T24細(xì)胞凋亡相關(guān)。
[Abstract]:Objective to down-regulate the expression of GSTP1 gene in human bladder cancer cell line T24 by using lentivirus expression system and to investigate its effect on the proliferation of T24 cells and its possible mechanism. Methods T24 cell lines with low expression of GSTP1 gene were established by lentivirus expression system, and the effects of down-regulation of GSTP1 expression on the proliferation of T24 cells were detected by CCK-8 assay and clone formation assay. Annexin V-FITC/PI staining and flow cytometry were used to detect the effect of down-regulation of GSTP1 expression on apoptosis of T 24 cells. Results compared with the untreated group and the control group, the proliferation of T24 cells in the GSTP1 interference group was significantly inhibited (P0.01). The results of apoptosis detection showed that the down-regulated expression of GSTP1 significantly promoted the apoptosis of T24 cells (P0.01). Conclusion the down-regulation of GSTP1 expression inhibits the proliferation of bladder cancer cell line T24, and its mechanism may be related to the promotion of T24 cell apoptosis.
