普羅布考對腎臟缺血再灌注損傷大鼠的保護(hù)作用及機制

【摘要】:目的觀察普羅布考對缺血再灌注大鼠腎臟氧化應(yīng)激損傷的保護(hù)作用,同時探討其作用機制。方法雄性SD大鼠30只,隨機分為假手術(shù)組(S組)、缺血再灌注組(IR組)、普羅布考治療組(P+IR組),每組10只。P+IR組大鼠每天用普羅布考(500mg/kg)灌胃,S組和IR組大鼠每天用等量溫開水灌胃。1周后制備腎缺血再灌注大鼠模型:3組大鼠都摘除右腎,IR組和P+IR組用無創(chuàng)動脈夾夾閉左側(cè)腎動脈,30min后松夾恢復(fù)血流再灌注。再灌注后S組和IR組每天溫開水灌胃,P+IR組每天普羅布考灌胃,連續(xù)1周。1周后處死所有大鼠,留取血及腎組織標(biāo)本,檢測大鼠各項血液生化指標(biāo),觀察腎組織病理學(xué)改變。結(jié)果與IR組相比,P+IR組血尿素氮(BUN)、血肌酐(SCr)、血脂水平明顯降低(均P0.05),血清和腎組織中SOD活性明顯上升、MDA含量明顯下降(均P0.05),腎組織病理損傷減輕,腎指數(shù)明顯改善(P0.05)。結(jié)論普羅布考能提高缺血再灌注損傷大鼠血及腎組織中SOD活性,改善腎功能,降低血脂水平,減輕腎組織病理損傷,發(fā)揮腎臟保護(hù)作用。
[Abstract]:Objective to observe the protective effect of Probucol on renal oxidative stress injury in ischemic reperfusion rats and to explore its mechanism. Methods Thirty male SD rats were randomly divided into three groups: sham operation group (S group), ischemia reperfusion group (IR group) and probucol treatment group (P IR group). 10. P IR rats in each group were treated with 500mg/kg daily. Rats in group S and group IR were perfused with the same amount of boiling water every day. 1 week later, the rat model of renal ischemia-reperfusion was established. The right kidney was excised in three groups, and the left renal artery was clamped by non-invasive artery in IR group and P IR group. The left renal artery was clamped by non-invasive artery in IR group and P IR group. After reperfusion, the rats in group S and group IR were fed with probucol daily with warm boiled water every day. All the rats were killed after 1 week. Blood and renal tissue samples were collected, and the blood biochemical indexes of the rats were detected. Renal histopathological changes were observed. Results compared with IR group, serum urea nitrogen (BUN), serum creatinine (SCr), serum lipid level (P 0.05), serum and renal SOD activity and MDA content were significantly decreased in, P IR group (P0.05). The pathological injury of renal tissue was alleviated and the renal index was significantly improved (P0.05). Conclusion Probucol can improve the activity of SOD in blood and renal tissue, improve renal function, decrease the level of blood lipids, alleviate the pathological injury of renal tissue and play a protective role in renal injury.
