RNA干擾ROR-γt表達對大鼠腎移植模型中Th17細胞功能和慢性排斥反應(yīng)的作用

【摘要】:目的探討RNA干擾Th17關(guān)鍵轉(zhuǎn)錄因子ROR-γt表達后,對大鼠腎移植慢性排斥模型中Th17細胞功能以及慢性排斥反應(yīng)的影響。方法構(gòu)建ROR-γt特異性干擾腺病毒,將其通過尾靜脈注入大鼠慢性排斥反應(yīng)模型,監(jiān)測大鼠血肌酐變化,酶聯(lián)免疫吸附試驗檢測Th17相關(guān)細胞因子IL-17、IL-6和TNF-γ改變。4周后檢測大鼠腎組織病理改變。結(jié)果成功構(gòu)建ROR-γt特異性干擾腺病毒pSES-HUS-ROR-γt。大鼠慢性排斥反應(yīng)模型尾靜脈注入pSES-HUS-ROR-γt,大鼠血清肌酐水平下降,IL-17、IL-6和TNF-γ水平下降,腎小球纖維化病變減輕。結(jié)論 pSES-HUS-ROR-γt特異性干擾腺病毒可以抑制Th17細胞功能,減輕腎移植慢性排斥反應(yīng)。
[Abstract]:Objective to investigate the effects of RNA interference on the expression of ROR- 緯 t, a key transcription factor of Th17, on the function of Th17 cells and chronic rejection in chronic renal allograft rejection model in rats. Methods ROR- 緯 t specific interfering adenovirus was constructed and injected into rat model of chronic rejection through tail vein. The changes of serum creatinine in rats were monitored, and the Th17 related cytokine IL-17, was detected by enzyme linked immunosorbent assay (Elisa). The changes of IL-6 and TNF- 緯 were detected after 4 weeks. Results ROR- 緯 t specific interfering adenovirus pSES-HUS-ROR- 緯 t was successfully constructed. PSES-HUS-ROR- 緯 t was injected into the caudal vein of chronic rejection model in rats. Serum creatinine level decreased, IL-17,IL-6 and TNF- 緯 levels decreased and glomerular fibrosis was alleviated. Conclusion pSES-HUS-ROR- 緯 t interference with adenovirus can inhibit the function of Th17 cells and alleviate the chronic rejection of renal transplantation.
