【摘要】：目的觀察調(diào)節(jié)小分子GTP結(jié)合蛋白Rab27表達(dá)對人膀胱癌細(xì)胞中細(xì)胞周期蛋白(Cyclin)、磷酸化黏著斑激酶抗體(p-FAK)、核因子κB抑制蛋白(p-IκB)表達(dá)的影響。方法取對數(shù)生長期人膀胱癌細(xì)胞系BIU-87(Rab27低表達(dá)細(xì)胞系)用Rab27質(zhì)粒轉(zhuǎn)染,人膀胱癌細(xì)胞系5637(Rab27高表達(dá)細(xì)胞系)用Rab27干擾質(zhì)粒轉(zhuǎn)染。采用Western blotting法檢測轉(zhuǎn)染前后BIU-87及5637細(xì)胞Rab27、Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB。結(jié)果與Rab27質(zhì)粒轉(zhuǎn)染前比較,轉(zhuǎn)染后BIU-87細(xì)胞Rab27的相對表達(dá)量升高,細(xì)胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB相對表達(dá)量均升高(P均0.05)。與Rab27干擾質(zhì)粒轉(zhuǎn)染前比較,5637細(xì)胞Rab27的相對表達(dá)量降低(P均0.05),細(xì)胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB相對表達(dá)量均降低(P均0.05)。結(jié)論轉(zhuǎn)染Rab27質(zhì)粒的BIU-87細(xì)胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB的表達(dá)升高。轉(zhuǎn)染Rab27干擾質(zhì)粒的5637細(xì)胞Cyclin D1、Cyclin E、p-FAK(Tyr925)、p-FAK(Tyr576/577)及p-IκB的表達(dá)降低。Rab27可能通過調(diào)節(jié)NF-κB、FAK信號通路的相關(guān)蛋白表達(dá)來影響膀胱癌細(xì)胞周期。
[Abstract]:Objective to investigate the effect of regulating the expression of small molecule GTP binding protein (Rab27) on the expression of cyclin (Cyclin), phosphorylated adhesion plaque kinase antibody (p-FAK) and nuclear factor 魏 B inhibitor protein (p-I 魏 B) in human bladder cancer cells. Methods Human bladder cancer cell line BIU-87 (Rab27 low expression cell line) in logarithmic phase was transfected with Rab27 plasmid, and human bladder cancer cell line 5637 (Rab27 overexpression cell line) was transfected with Rab27 interference plasmid. Western blotting assay was used to detect Rab27,Cyclin D1Cyclin Ep-FAK (Tyr925), p-FAK (Tyr576/577) and p-I 魏 B in BIU-87 and 5637 cells before and after transfection. Results compared with those before transfection of Rab27 plasmid, the relative expression of Rab27 in BIU-87 cells was increased after transfection, and the relative expression of Tyr925, p-FAK (Tyr576/577) and p-I 魏 B were increased in Cyclin D1Cyclin E-FAK (Tyr925), p-FAK (Tyr576/577) and p-I 魏 B (P0. 05). Compared with that before transfection with Rab27 interference plasmid, the relative expression of Rab27 in 5637 cells was significantly lower than that before transfection (P 0.05), and the relative expression of Cyclin D1Cyclin E-FAK (Tyr925), p-FAK (Tyr576/577) and p-I 魏 B were all decreased (P 0.05). Conclusion the expression of p-FAK (Tyr925), p-FAK (Tyr576/577) and p-I 魏 B in BIU-87 cells transfected with Rab27 plasmid were increased. The expression of p-FAK (Tyr925), p-FAK (Tyr576/577) and p-I 魏 B in 5637 cells transfected with Rab27 interference plasmid was decreased. Rab27 may affect the cell cycle of bladder cancer by regulating the expression of the related proteins in NF- 魏 B pFAK signaling pathway.
