淫羊藿苷聯(lián)合胚胎干細胞對造影劑腎病大鼠模型腎上皮細胞凋亡的影響

【摘要】:目的:探討淫羊藿苷聯(lián)合胚胎干細胞對造影劑腎病(CIN)大鼠模型腎上皮細胞的作用及其可能的機制,為臨床上淫羊藿苷聯(lián)合應用提供實驗依據(jù)。方法:選取SD大鼠50只,隨機分為對照組、模型組、淫羊藿苷組、干細胞組和聯(lián)合給藥組,每組10只,除對照組外其他各組大鼠尾靜脈注射吲哚美辛(INDO)、N-硝基-LO-精氨酸甲酯(L-NAME)和泛影葡胺建立CIN大鼠模型。檢測造模前后血肌酐(Scr)值;采用半定量RT-PCR和Western blotting法檢測細胞凋亡相關因子p38MAPK和caspase 3mRNA和蛋白的表達,流式細胞術定量檢測腎上皮細胞凋亡率。結果:Scr測定,注射造影劑48h后,大鼠Scr值明顯升高,提示造模成功。與模型組比較,聯(lián)合給藥組大鼠Scr值明顯下降(P0.05)。與模型組比較,淫羊藿苷組、干細胞組和聯(lián)合給藥組大鼠p38MAPK和caspase-3mRNA和蛋白表達水平降低(P0.05);聯(lián)合給藥組大鼠腎上皮細胞凋亡率明顯低于淫羊藿苷組和干細胞組(P0.05)。結論:聯(lián)合給藥對CIN模型大鼠的腎上皮細胞有保護作用,其作用機制可能與抑制細胞凋亡通路中相關因子有關。
[Abstract]:Objective: to investigate the effect of icariin combined with embryonic stem cells on renal epithelial cells in (CIN) rats model of contrast medium nephropathy and its possible mechanism, and to provide experimental evidence for clinical application of icariin. Methods: fifty SD rats were randomly divided into control group, model group, icariin group, stem cell group and combined administration group, with 10 rats in each group. The rat model of CIN was established by injecting indomethacin (INDO) (N-nitro-L-arginine methyl ester) (L-NAME) and meglumine diatrizoate into the tail vein of rats except the control group. The levels of serum creatinine (Scr), the expression of apoptosis-related factors p38MAPK, caspase 3mRNA and protein were detected by semi-quantitative RT-PCR and Western blotting, and the apoptosis rate of renal epithelial cells was quantitatively detected by flow cytometry. Results the Scr value of rats increased significantly 48 hours after injection of contrast agent, indicating that the model was successful. Compared with the model group, the Scr value of the combined administration group decreased significantly (P0.05). Compared with model group, the expression of p38MAPK and caspase-3mRNA and protein in Icariin group, stem cell group and combined administration group were significantly lower than those in Icariin group and stem cell group (P0.05), while the apoptosis rate of renal epithelial cells in combined administration group was significantly lower than that in Icariin group and stem cell group (P0.05). Conclusion: combined administration of drugs has protective effect on renal epithelial cells in CIN model rats, and its mechanism may be related to the inhibition of factors related to apoptosis pathway.
