轉(zhuǎn)移性腎細(xì)胞癌中RSK4、CD44及MMP-9的表達(dá)及意義

【摘要】:目的檢測核糖體S6蛋白激酶4(ribosomal S6 protein kinase 4,RSK4)、CD44和MMP-9在原發(fā)性腎細(xì)胞癌(primary renal cell carcinoma,pRCC)及轉(zhuǎn)移性腎細(xì)胞癌(metastatic renal cell carcinoma,mRCC)中的表達(dá),探討三者表達(dá)水平及與臨床病理特征的相關(guān)性。方法采用免疫組化En Vision法檢測52例pRCC及48例mRCC中RSK4、MMP-9和CD44蛋白的表達(dá),并分析三者表達(dá)的相關(guān)性及與臨床病理特征的關(guān)系。結(jié)果 48例mRCC中RSK4、CD44及MMP-9的陽性率分別為75%(36/48)、68.75%(33/48)及91.7%(44/48),而三者在52例pRCC中的陽性率分別為44.2%(23/52)、34.6%(18/52)及69.2%(36/52)。RSK4、CD44及MMP-9在mRCC中的表達(dá)高于pRCC(PRSK4=0.002;PMMP-9=0.002;PCD44=0.001);RSK4、CD44及MMP-9表達(dá)與mRCC患者年齡、性別和腫瘤Fuhrman分級、轉(zhuǎn)移部位之間未見明確相關(guān)性(P0.05)。在mRCC中,三者之間的表達(dá)呈正相關(guān)(P=0.008);兩兩相關(guān)分析發(fā)現(xiàn)RSK4與CD44(P=0.019)、MMP-9與CD44(P=0.05)的表達(dá)之間存在正相關(guān),而RSK4與MMP-9(P=1.00)未見明確相關(guān)性。結(jié)論 RSK4、CD44及MMP-9在mRCC中的表達(dá)高于pRCC,提示三者可能介導(dǎo)腎細(xì)胞癌(renal cell carcinoma,RCC)的轉(zhuǎn)移,而其具體作用機(jī)制尚有待進(jìn)一步研究。
