G蛋白信號轉導調節(jié)因子4在腎癌組織中的表達及意義

【摘要】:目的探討腎細胞癌(RCC)組織中G蛋白信號轉導調節(jié)因子4(RGS4)的表達變化及其與RCC患者臨床病理特征的關系。方法收集120例RCC患者手術切除的癌組織及癌旁組織,采用免疫組化染色及Western blot法檢測RGS4蛋白表達,RT-PCR法檢測RGS4 mRNA表達,分析不同臨床病理特征的RCC患者癌組織RGS4蛋白表達變化。結果免疫組化染色示,RGS4特異性表達于腎細胞的細胞質中,在癌旁組織中為淺黃色,在癌組織中為深黃色。癌旁組織、RCC癌組織中RGS4蛋白表達陽性率分別為24.2%、37.5%,RCC癌組織中RGS4蛋白表達陽性率高于癌旁組織(P0.05)。癌旁組織、RCC癌組織RGS4蛋白表達量分別為0.66±0.03、1.02±0.03,RCC癌組織RGS4蛋白表達量高于癌旁組織(P0.05)。癌旁組織、RCC癌組織RGS4 mRNA表達量分別為0.93±0.04、1.67±0.04,RCC癌組織RGS4 mRNA表達量高于癌旁組織(P0.05)。中度及重度浸潤的RCC患者RGS4表達率顯著高于輕度浸潤的患者(P0.05)。結論 RCC癌組織中RGS4表達增高,并隨浸潤程度增加表達率顯著升高,參與RCC的發(fā)生發(fā)展。
[Abstract]:Objective to investigate the expression of G protein signal transduction regulatory factor 4 (RGS4) in renal cell carcinoma (RCC) and its relationship with the clinicopathological features of RCC. Methods the expression of RGS4 mRNA was detected by immunohistochemical staining and Western blot method in 120 patients with RCC. The expression of RGS4 protein was detected by RT-PCR in RCC patients with different clinicopathological features. The expression of RGS4 protein was detected by immunohistochemical staining and RT-PCR. The changes of RGS4 protein expression in the tissues of RCC patients with different clinicopathological features were analyzed. Results Immunohistochemical staining showed that RGS4 was specifically expressed in the cytoplasm of renal cells, which was yellowish in paracancerous tissues and dark yellow in cancerous tissues. The positive rate of RGS4 protein expression in paracancerous tissues was higher than that in paracancerous tissues (P 0.05). The expression of RGS4 protein in paracancerous tissues was higher than that in paracancerous tissues (P 0.05). The expression of RGS4 mRNA in paracancerous tissues was higher than that in paracancerous tissues (0. 93 鹵0. 04) and 1. 67 鹵0. 04 respectively (P0.05). The expression rate of RGS4 in RCC patients with moderate and severe infiltration was significantly higher than that with mild invasion (P0.05). Conclusion the expression of RGS4 in RCC carcinoma is increased and the expression rate is increased with the degree of invasion, which is involved in the occurrence and development of RCC.
