【摘要】：目的初步探討擴增活化的自體淋巴細胞(expanded activated autologous lymphocytes,EAAL)對裸鼠腎母細胞瘤(Wilms' tumor,WT)移植瘤生長的影響。方法實驗中腫瘤組織來源于術(shù)前穿刺活檢病理證實腎母細胞瘤的患者,術(shù)中切取部分腫瘤組織并制備保存。將2-3塊約1mm*1mm*1mm大小的腎母細胞瘤腫瘤組織種植在右腋窩皮下,并將傷口嚴密縫合,隨后將裸鼠放回籠內(nèi)飼養(yǎng)并嚴密觀察裸鼠的生長變化。小鼠瘤體模型建立后,選取其中15只裸鼠隨機分為3組。不同處理組經(jīng)尾靜脈分別輸注等量且不同劑量的EAAL細胞懸浮液和等量生理鹽水,根據(jù)對每組裸鼠不同的處理方式,分為低劑量組、高劑量組和陰性對照組。每組裸鼠經(jīng)8次注射,末次注射l周后頸椎脫臼法處死裸鼠,剝離腫瘤組織并稱重,用游標卡尺準確測量腫瘤直徑。采用配對t檢驗比較每組裸鼠處理前后的差異變化。單因素方差分析比較不同處理組間的差異變化。P0.05確定為差異有統(tǒng)計學(xué)意義。結(jié)果實驗結(jié)果表明:注射EAAL細胞和生理鹽水后,比較各組間裸鼠腫瘤直徑變化有差異,且低、高劑量組較陰性對照組裸鼠腫瘤直徑縮小明顯(0.97±0.10cm、0.64±0.07 cm vs 1.33±0.09 cm;P0.05)。瘤體質(zhì)量方面,低、高劑量組亦較陰性對照組裸鼠明顯減輕(0.96±0.09g、0.62±0.05g vs 1.20±0.11g;P0.05)。比較注射EAAL細胞兩組,高劑量組比低劑量組對腫瘤的抑制作用明顯。結(jié)論EAAL細胞治療能夠影響裸鼠腎母細胞瘤移植瘤的生長,且可能對裸鼠腎母細胞瘤移植瘤的生長有抑制作用。
[Abstract]:Objective to investigate the effect of amplified activated autologous lymphocyte (expanded activated autologous lymphocytes) on the growth of xenografts in nude mice with Wilms' tumor. Methods tumor tissues were obtained from patients with nephroblastoma confirmed by biopsy before operation. Two or three pieces of tumor tissue about the size of 1mm*1mm*1mm were implanted in the right armpit subcutaneously and the wound was sutured closely. Then the nude mice were reared in cage and the growth of nude mice was observed closely. After the establishment of mouse tumor model, 15 nude mice were randomly divided into 3 groups. The EAAL cell suspensions and the same amount of normal saline were infused into the caudal vein in different groups. According to the different treatment methods of each group, they were divided into three groups: low dose group, high dose group and negative control group. The nude mice in each group were injected for 8 times and the cervical spine dislocations were performed after the last injection for 1 week. The tumor tissues were removed and weighed. The tumor diameter was accurately measured with Vernier caliper. Paired t test was used to compare the difference before and after treatment in each group of nude mice. Univariate ANOVA compared the differences between different treatment groups. P05 was determined to be the difference was statistically significant. Results the results showed that the diameter of tumor in the high dose group was significantly smaller than that in the negative control group (0.97 鹵0.10 cm ~ (-1) 鹵0.07 cm vs 1.33 鹵0.09 cm ~ (-1) P 0.05) after injection of EAAL cells and normal saline. The tumor diameter of the nude mice was lower than that of the control group (0.97 鹵0.10 cm ~ (-1) vs 1.33 鹵0.09 cm ~ (-1). The tumor mass in the low and high dose group was significantly reduced compared with that in the negative control group (0.96 鹵0.09 g / kg vs 1.20 鹵0.11 g / g, P 0.05). Compared with EAAL cells injection group, the high dose group was more effective than the low dose group on tumor inhibition. Conclusion EAAL cell therapy can affect the growth of xenografts of nephroblastoma in nude mice and may inhibit the growth of xenografts of nephroblastoma in nude mice.
