【摘要】：目的探討鹽霉素對(duì)前列腺癌DU145干細(xì)胞的影響及其可能的機(jī)制,為鹽霉素的臨床應(yīng)用提供理論依據(jù)。方法鹽霉素處理DU145細(xì)胞,以ALDH為腫瘤干細(xì)胞標(biāo)記物,用流式細(xì)胞儀檢測(cè)處理后ALDH陽性的DU145干細(xì)胞的百分比。鹽霉素處理DU145細(xì)胞后,用Western Blot法檢測(cè)mTOR信號(hào)通路相關(guān)蛋白m-TOR、p-70s6k、p-p70s6、p-s6等的表達(dá)變化。鹽霉素結(jié)合mTOR信號(hào)通路抑制劑雷帕霉素處理DU145細(xì)胞,利用流式細(xì)儀檢測(cè)ALDH陽性的DU145干細(xì)胞百分比。結(jié)果鹽霉素顯著性抑制ALDH陽性的DU145干細(xì)胞(抑制率為77.78%),是紫杉醇的2倍(抑制率為38.64%)。鹽霉素抑制mTOR信號(hào)通路相關(guān)分子m-TOR、p-70s6k、p-p70s6、p-s6等蛋白的表達(dá),具有時(shí)間依賴性和劑量依賴性。鹽霉素結(jié)合雷帕霉素能夠抑制ALDH陽性的DU145干細(xì)胞比例(抑制率為77.95%)。結(jié)論鹽霉素可能通過抑制mTOR通路信號(hào)來發(fā)揮抑制腫瘤干細(xì)胞的作用。
[Abstract]:Objective to investigate the effect of salinomycin on prostate cancer DU145 stem cells and its possible mechanism, and to provide theoretical basis for the clinical application of salinomycin. Methods DU145 cells were treated with salinomycin and ALDH was used as the marker of tumor stem cells. Flow cytometry was used to detect the percentage of ALDH-positive DU145 stem cells. After treated with salinomycin, the expression of mTOR signaling pathway related protein m-TORp-70s6kN, p-p70s6, p-s6 and so on was detected by Western Blot. DU145 cells were treated with salinomycin combined with rapamycin, an inhibitor of mTOR signaling pathway, and the percentage of ALDH positive DU145 stem cells was detected by flow cytometry. Results Salinomycin significantly inhibited ALDH-positive DU145 stem cells (inhibition rate was 77.78%, twice as much as paclitaxel) (inhibition rate was 38.64%). Salinomycin inhibited the expression of mTOR signaling pathway related molecules m-TOR-p-70s6knp-p70s6p-s6 in a time-and dose-dependent manner. Salinomycin combined with rapamycin could inhibit the percentage of ALDH-positive DU145 stem cells (inhibition rate was 77.95%). Conclusion Salinomycin may inhibit tumor stem cells by inhibiting the signal of mTOR pathway.
