大鼠睪丸開放活檢導(dǎo)致生精小管形態(tài)改變

【摘要】:研究背景及目的:筆者以前參與的研究表明,成年大鼠睪丸開放活檢,導(dǎo)致了生精細(xì)胞團(tuán)向生精小管腔內(nèi)突出的形態(tài)改變。本文擬進(jìn)一步研究確定大鼠睪丸活檢導(dǎo)致的生精小管形態(tài)改變,同時(shí)利用連續(xù)切片觀察生精小管的形態(tài)以探討活檢所致形態(tài)改變的形成機(jī)制。材料與方法:14只成年大鼠麻醉后在一側(cè)睪丸作切口擠出組織塊,浸潤(rùn)固定整個(gè)睪丸后切取擠出的組織塊(未游離活檢塊)。另8只成年大鼠麻醉后在一側(cè)睪丸作切口擠出組織塊并立即切下,然后固定(游離活檢塊)。這22只動(dòng)物的另一側(cè)睪丸,浸潤(rùn)固定后切取睪丸組織塊(原位組織塊)。所有組織塊都用甲基丙烯酸樹脂包埋,從每個(gè)包埋塊切取1張切片(25μm厚)。另外,從每個(gè)游離活檢塊再切平均46張連續(xù)切片。所有切片用過碘酸-席夫試劑和蘇木精染色,然后進(jìn)行組織學(xué)觀察分析。結(jié)果:與原位組織塊相比,未游離活檢塊的生精小管基本正常,而在切自游離活檢塊的切片上,有約一半的生精小管輪廓腔內(nèi)可見單獨(dú)的生精細(xì)胞團(tuán),這些生精細(xì)胞團(tuán)幾乎都是由精母細(xì)胞及精子細(xì)胞組成。連續(xù)切片觀察提示,這些生精細(xì)胞團(tuán)來(lái)自某處的生精小管壁,開放活檢過程中被擠出或突向鄰近的生精小管腔。結(jié)論:成熟睪丸開放活檢可導(dǎo)致生精細(xì)胞團(tuán)從生精上皮脫落,突向管腔,這可能是活檢時(shí)游離組織塊內(nèi)生精小管的局部收縮所致。
[Abstract]:Background and objective: open biopsy of testis in adult rats has resulted in morphological changes of spermatogenic cell clusters protruding into seminiferous tubules. This paper intends to further study and determine the morphological changes of seminiferous tubules induced by testicular biopsy in rats, and observe the morphology of seminiferous tubules by serial sections in order to explore the formation mechanism of the morphological changes caused by testicular biopsies. Materials and methods 14 adult rats were anesthetized and extruded tissue mass (not free biopsy mass) in one testicle was cut out by infiltrating and fixing the whole testis. The other 8 adult rats were anesthetized and cut out the tissue mass through a unilateral testicular incision and then fixed (free biopsy mass). On the other side of the testis of the 22 animals, the testicular mass (in situ tissue mass) was removed after invasion and fixation. All tissue blocks were encapsulated with methacrylic resin, and a slice of 25 渭 m thick was cut from each embedding block. In addition, an average of 46 consecutive sections were resliced from each free biopsy mass. All sections were stained with iodate-Schiff reagent and hematoxylin. Results: compared with the in situ tissue mass, the seminiferous tubules without free biopsy mass were basically normal, but in the section cut from the free biopsy mass, about half of the seminiferous tubules could be seen in the contours of seminiferous tubules. These spermatogenic cell clusters are almost composed of spermatocytes and sperm cells. Serial microscopic observations suggest that these spermatogenic cell clusters come from somewhere in the seminiferous tubule wall and are extruded or protruded into the adjacent spermatogenic tubules during open biopsy. Conclusion: open biopsy of mature testis can cause spermatogenic cell mass to fall off from spermatogenic epithelium and protrude to lumen, which may be due to the local contraction of endophytic tubules in free tissue mass during biopsy.
