NGF和CgA對(duì)RWPE-1細(xì)胞內(nèi)鈣離子濃度變化的影響及其在慢性前列腺炎中的作用

【摘要】:目的探討神經(jīng)生長(zhǎng)因子(NGF)和嗜鉻顆粒蛋白A(CgA)處理后前列腺上皮細(xì)胞系RWPE-1細(xì)胞內(nèi)鈣離子濃度([Ca~(2+)]_i)的差異及NGF和CgA在慢性前列腺炎(CP)發(fā)病中的作用。方法不同濃度的NGF和CgA(50、200 ng/ml)刺激RWPE-1細(xì)胞,用激光共聚焦顯微鏡觀(guān)察不同濃度的NGF和CgA刺激后細(xì)胞內(nèi)[Ca~(2+)]_i變化,加入TRPV1受體拮抗劑后繼續(xù)觀(guān)察細(xì)胞內(nèi)[Ca~(2+)]_i變化。然后,將正常細(xì)胞外液換成D-PBS,再加入NGF和CgA觀(guān)察細(xì)胞內(nèi)[Ca~(2+)]_i變化。結(jié)果 50 ng/ml NGF和CgA刺激細(xì)胞后,細(xì)胞內(nèi)[Ca~(2+)]_i未見(jiàn)明顯變化。200 ng/ml NGF和CgA刺激細(xì)胞后,細(xì)胞內(nèi)[Ca~(2+)]_i明顯增加,加入TRPV1受體拮抗劑后,細(xì)胞內(nèi)[Ca~(2+)]_i未見(jiàn)明顯變化。將正常細(xì)胞外液換成D-PBS,用NGF和CgA刺激后,細(xì)胞內(nèi)[Ca~(2+)]_i未見(jiàn)明顯改變。結(jié)論CP的發(fā)生可能是通過(guò)分泌和釋放NGF和CgA,引起前列腺上皮細(xì)胞膜TRPV1通道開(kāi)放,促使細(xì)胞內(nèi)鈣離子濃度增加,導(dǎo)致前列腺上皮分泌功能下降所致。
[Abstract]:Objective to investigate the difference of intracellular calcium concentration ([Caan2] I] in prostatic epithelial cell line RWPE-1 treated with NGF and CgA and the role of NGF and CGA in the pathogenesis of chronic prostatitis. Methods RWPE-1 cells were stimulated with different concentrations of NGF and CgA150 ng / ml. The changes of [Caan2] I in RWPE-1 cells were observed by confocal laser microscopy, and the intracellular [Caan2] I changes were observed after adding TRPV1 receptor antagonist to RWPE-1 cells. Then, the normal extracellular fluid was replaced with D-PBSand then NGF and CGA were added to observe the changes of [CaO2] I in the cells. Results after stimulation with 50 ng/ml NGF and CGA, no significant changes were observed in [Cajian2] in the cells. After stimulation with CGA and CGA, the intracellular [Cajian2] I was significantly increased, but no significant changes were observed in the [Cajian2] I after the addition of TRPV1 receptor antagonist. After the normal extracellular fluid was replaced with D-PBSand stimulated by NGF and CGA, the intracellular [Cajian2] had no obvious change. Conclusion CP may be caused by the secretion and release of NGF and CgA, which may lead to the opening of TRPV1 channel of prostate epithelial cell membrane and the increase of intracellular Ca ~ (2 +) concentration, resulting in the decrease of secretory function of prostatic epithelium.
