【摘要】：目的通過(guò)觀察選擇性雄激素受體調(diào)節(jié)劑對(duì)遲發(fā)性腺功能減退癥(LOH)大鼠的作用,為治療LOH提供理論依據(jù)。 方法將30只符合標(biāo)準(zhǔn)的老年雄性Wistar大鼠作為L(zhǎng)OH動(dòng)物模型,并隨機(jī)分成3組：對(duì)照組(A)、DHT組(B)、Sarm組(C),分別給予等計(jì)量的空白溶劑、雄激素及選擇性雄激素受體調(diào)節(jié)劑(Sarm)進(jìn)行干預(yù),實(shí)驗(yàn)4周后觀察各組大鼠LOH癥狀的改善情況并評(píng)分,記錄大鼠懸尾及強(qiáng)迫游泳時(shí)間,檢測(cè)血清游離睪酮的水平,計(jì)算前列腺指數(shù)。 結(jié)果干預(yù)4周后A、B、C三組大鼠LOH癥狀評(píng)分分別為(18.20±1.87)、(30.00±2.49)、(35.20±1.87),C組優(yōu)于B組,B組優(yōu)于A組,差異有統(tǒng)計(jì)學(xué)意義；懸尾不動(dòng)時(shí)間(s)分別為(196.17±10.57)、(167.29±2.15)、(160.67±6.92),C組短于B組,B組短于A組,差異有統(tǒng)計(jì)學(xué)意義；強(qiáng)迫游泳時(shí)間(min)分別為(16.33±1.16)、(24.50±1.26)、(30.43±0.71),C組長(zhǎng)于B組,B組長(zhǎng)于A組,差異有統(tǒng)計(jì)學(xué)意義;血清游離睪酮(pg/mL)分別為(2.248±0.305)、(1.088±0.258)、(2.526±0.283),B組低于A組和C組,差異有統(tǒng)計(jì)學(xué)意義；前列腺指數(shù)(%)分別為(0.321±0.008)、(0.419±0.015)、(0.172±0.007),B組高于A組而C組低于A組,各組間差異有統(tǒng)計(jì)學(xué)意義(P0.01)。 結(jié)論Sarm可通過(guò)其與雄激素受體高度親和力及選擇性調(diào)節(jié)雄激素受體表達(dá),明顯改善LOH模型大鼠的LOH癥狀,有效抑制前列腺組織的增生,不影響自身睪酮的分泌。
[Abstract]:Objective to observe the effect of selective androgen receptor modulator on delayed hypogonadism in rats and to provide a theoretical basis for the treatment of LOH. Methods Thirty old male Wistar rats were randomly divided into three groups: control group (control group) and control group. The control group was treated with equal dose of blank solvent, androgen and selective androgen receptor modulator. After 4 weeks of experiment, the improvement of LOH symptoms and scores were observed, the time of tail suspension and forced swimming were recorded, the level of serum free testosterone was measured, and the prostate index was calculated. Results after 4 weeks of intervention, the scores of LOH symptoms in group C were 18.20 鹵1.87, 30.00 鹵2.49 and 35.20 鹵1.87, respectively, which were significantly better than those in group B, and the time of suspension immobility was 196.17 鹵10.577.29 鹵2.150.67 鹵6.92, respectively, which was significantly shorter than that in group B (P < 0.05). The time of forced swimming was 16.33 鹵1.16, respectively (24.50 鹵1.26). Group C was longer than group B (30.43 鹵0.71) than group B, the difference was statistically significant, the serum free testosterone pg / mL was 2.248 鹵0.305 and 1.088 鹵0.2588 鹵0.2583, respectively, and the difference was statistically significant. The prostatic index was 0.321 鹵0.008 (0.419 鹵0.015) in group B was higher than that in group A (0.172 鹵0.007), but lower in group C than in group A (P 0.01). Conclusion Sarm can regulate androgen receptor expression through its high affinity and selectivity with androgen receptor, improve LOH symptoms of LOH model rats, inhibit prostatic hyperplasia effectively, and do not affect the secretion of self testosterone.
