白皮杉醇抑制前列腺癌細(xì)胞株增殖、遷移與侵襲

【摘要】:目的:觀察白皮杉醇對(duì)前列腺癌細(xì)胞株DU145的細(xì)胞活力、遷移與侵襲能力的作用并初步討論其機(jī)制。方法:CCK-8法檢測(cè)不同濃度白皮杉醇(0、5、10、20、40和80μmol/L)作用不同時(shí)間(12、24、36和48 h)對(duì)DU145細(xì)胞活力的影響;劃痕實(shí)驗(yàn)與Transwell小室法分別檢測(cè)白皮杉醇對(duì)DU145細(xì)胞遷移與侵襲能力的影響;Western blot法檢測(cè)p-JAK2與p-STAT3的蛋白水平。結(jié)果:CCK-8法結(jié)果顯示白皮杉醇呈濃度依賴性抑制DU145細(xì)胞的活力;給予白皮杉醇干預(yù)后,細(xì)胞遷移數(shù)和侵襲數(shù)顯著減少,p-JAK2與p-STAT3蛋白水平顯著下降。結(jié)論:白皮杉醇呈濃度依賴性抑制前列腺癌細(xì)胞的生長(zhǎng),并具有抑制細(xì)胞遷移與侵襲的作用,其機(jī)制可能與抑制JAK2/STAT3信號(hào)通路有關(guān)。
[Abstract]:Aim: to observe the effect of Huperzine on the viability, migration and invasion of prostate cancer cell line DU145 and to discuss its mechanism. Methods the effects of different concentrations of Huperzine 10101040 and 80 渭 mol / L on the activity of DU145 cells at different time points (124g / L and 48h) were detected by the method of 10% CCK-8. The effects of taxol on the migration and invasion of DU145 cells were detected by scratch assay and Transwell chamber assay. Western blot assay was used to detect the protein levels of p-JAK2 and p-STAT3. Results the cell viability of DU145 cells was inhibited in a concentration-dependent manner by% CCK-8 method, and the number of migration and invasion decreased significantly after treatment with Huperzine, and the levels of p-JAK2 and p-STAT3 protein decreased significantly. Conclusion: Huperzine inhibited the growth of prostate cancer cells in a concentration-dependent manner and inhibited the migration and invasion of prostate cancer cells. The mechanism may be related to the inhibition of JAK2/STAT3 signaling pathway.
