KLF5和TAZ在膀胱癌組織中的表達及臨床意義

【摘要】:目的:膀胱癌在泌尿系統(tǒng)腫瘤中的發(fā)病率和死亡率均居首位,具有難檢測、易復(fù)發(fā)、預(yù)后差等特點。膀胱癌的發(fā)病是多因素、多階段相互作用的復(fù)雜過程,其發(fā)病機制迄今尚未明確,因此仍缺乏有效的診斷、預(yù)防和治療措施。KLF5是近年發(fā)現(xiàn)的一種鋅指蛋白轉(zhuǎn)錄因子,能夠通過抑制或激活目的基因的轉(zhuǎn)錄調(diào)控基因表達,在細胞增殖、分化、凋亡和腫瘤的發(fā)生發(fā)展中都發(fā)揮重要作用。TAZ是一種轉(zhuǎn)錄共激活因子,能夠與多種調(diào)控因子相互作用,促進癌細胞的增殖和腫瘤的發(fā)生,KLF5轉(zhuǎn)錄因子可能與TAZ的WW結(jié)構(gòu)域相互作用促進癌細胞的增殖和腫瘤的形成。本研究通過檢測KLF5和TAZ在膀胱癌及正常膀胱組織中的表達,分析其與膀胱癌分化程度、病理分級、TNM分期等臨床病理參數(shù)的相關(guān)性以及兩者之間的相關(guān)性。 方法:應(yīng)用免疫組織化學(xué)方法檢測46例膀胱癌及10例正常膀胱組織中KLF5和TAZ的表達水平,分析兩者與膀胱癌分化程度、病理分級TNM分期等臨床病理參數(shù)的相關(guān)性以及兩種轉(zhuǎn)錄調(diào)控因子之間的相關(guān)性。 結(jié)果:膀胱癌組織中KLF5和TAZ的陽性表達率都高于正常膀胱組織,膀胱癌組織中KLF5的陽性表達率為78.3%(36/46),TAZ的陽性表達率為76.1%(35/46),與正常膀胱組織中的KLF5和TAZ表達量相比,都具有顯著性差異(P0.05)。KLF5和TAZ蛋白表達與腫瘤的分化程度(P=0.023;P=0.008)和病理分級(P=0.025;P=0.016)顯著相關(guān),與性別、病理TNM分期和腫瘤轉(zhuǎn)移無顯著相關(guān)性。 結(jié)論:膀胱癌組織中的TAZ和KLF5的表達上調(diào)與腫瘤的生長、分化、浸潤和預(yù)后都密切相關(guān),表明TAZ和KLF5是膀胱癌發(fā)生發(fā)展和患者預(yù)后優(yōu)劣的重要標(biāo)志物,能夠為膀胱癌的發(fā)生和癌變提供一個診斷指標(biāo),為今后的臨床研究提供重要的理論依據(jù)。
[Abstract]:Objective: the incidence and mortality of bladder cancer are the first in urological tumor. It has the characteristics of difficult detection, recurrence and poor prognosis. The pathogenesis of bladder cancer is a complex process of multiple factors and multistage interaction. The pathogenesis of bladder cancer is not clear so far. Therefore, the prevention and treatment measures of.KLF5 are still found in recent years. A zinc finger protein transcription factor, which can play an important role in cell proliferation, differentiation, apoptosis and tumor development by inhibiting or activating transcriptional regulation of gene expression in the target gene..TAZ is a transcriptional co activating factor, which can interact with a variety of regulators to promote the proliferation of cancer cells and the occurrence of tumors, KLF The 5 transcriptional factor may interact with the WW domain of TAZ to promote the proliferation of cancer cells and the formation of tumor. This study examined the expression of KLF5 and TAZ in bladder cancer and normal bladder tissue, and analyzed the correlation with the degree of differentiation, pathological grade, TNM staging and other clinicopathological parameters, and the correlation between them.
Methods: the expression of KLF5 and TAZ in 46 cases of bladder cancer and 10 normal bladder tissues was detected by immunohistochemistry. The correlation between the differentiation degree of bladder cancer, the clinicopathological parameters, such as the pathological grade TNM staging, and the correlation between the two kinds of transcription factors were analyzed.
Results: the positive expression rate of KLF5 and TAZ in bladder cancer tissues is higher than that of normal bladder tissue. The positive expression rate of KLF5 in bladder cancer tissues is 78.3% (36/46), and the positive expression rate of TAZ is 76.1% (35/46). Compared with the expression of KLF5 and TAZ in normal bladder tissue, there are significant differences (P0.05).KLF5 and TAZ protein expression and tumor score. The degree of P=0.023 (P=0.008) and pathological grading (P=0.025; P=0.016) were significantly correlated with gender, pathological TNM stage and tumor metastasis.
Conclusion: the up-regulated expression of TAZ and KLF5 in bladder cancer tissues is closely related to the growth, differentiation, infiltration and prognosis of the tumor. It shows that TAZ and KLF5 are important markers for the development of bladder cancer and the prognosis of the patients. It can provide a diagnostic index for the occurrence and canceration of bladder cancer and provide an important theory for future clinical research. Basis.
