骨橋蛋白與特發(fā)性草酸鈣結(jié)石的相關(guān)性研究

【摘要】:目的: 通過(guò)觀察特發(fā)性草酸鈣結(jié)石(idiopathic calcium oxalate stone,ICS)患者腎組織中骨橋蛋白(osteopontin,OPN)mRNA和蛋白的表達(dá)水平及尿液中OPN濃度的變化,探討OPN在ICS形成中的作用。 方法: 選取我院2013年1月至2014年1月期間因ICS致患腎無(wú)功能行腎切除術(shù)的34例腎組織標(biāo)本為實(shí)驗(yàn)組,其中男性16例,女性18例,年齡13歲~73歲,平均50.7歲;收集同期行根治性腎切除術(shù)患者遠(yuǎn)癌正常腎組織標(biāo)本38例作為對(duì)照組,其中男性16例,女性22例,年齡40歲~77歲,平均50.1歲。收集實(shí)驗(yàn)組和對(duì)照組患者尿液標(biāo)本及19例正常人尿液標(biāo)本(正常組)。采用Real-timeRT-PCR法檢測(cè)兩組腎組織中OPN mRNA的表達(dá);Western blot法檢測(cè)兩組腎組織中OPN蛋白的表達(dá)情況;ELISA法測(cè)定三組尿液中OPN濃度。統(tǒng)計(jì)學(xué)數(shù)據(jù)用x±s表示,用SPSS14.0軟件進(jìn)行統(tǒng)計(jì)學(xué)處理,兩組數(shù)據(jù)比較統(tǒng)計(jì)學(xué)分析采用兩個(gè)獨(dú)立樣本的t檢驗(yàn),三組數(shù)據(jù)比較統(tǒng)計(jì)學(xué)分析采用單因素方差分析。 結(jié)果: 實(shí)驗(yàn)組與對(duì)照組腎組織OPN mRNA相對(duì)表達(dá)量ΔCT分別為2.04±1.20、2.59±0.91,差異有統(tǒng)計(jì)學(xué)意義(P0.05);OPN蛋白的相對(duì)表達(dá)量分別為0.776±0.071、0.449±0.085,差異有統(tǒng)計(jì)學(xué)意義(P 0.05);實(shí)驗(yàn)組、對(duì)照組、正常組尿液中OPN蛋白濃度分別為1.052±0.229、1.579±0.256、1.498±0.155,實(shí)驗(yàn)組和對(duì)照組、實(shí)驗(yàn)組與正常組相比,差異有統(tǒng)計(jì)學(xué)意義(P0.05),對(duì)照組與正常組相比,,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論: ICS患者腎小管上皮細(xì)胞OPN mRNA和蛋白的過(guò)度表達(dá)及其尿液中OPN濃度的降低可能是腎結(jié)石形成的重要因素。
[Abstract]:Objective: To investigate the role of OPN in the formation of calcium oxalate by observing the expression level of osteopontinin in renal tissue and the concentration of OPN in urine in patients with idiopathic calcium oxalate stone (idiopathic calcium oxalate stone). Methods: From January 2013 to January 2014, 34 specimens of renal tissue were selected as experimental group, including 16 males and 18 females, aged from 13 to 73 years (mean 50.7 years). 38 cases of normal renal tissue of distal carcinoma were collected as control group, including 16 males and 22 females, aged from 40 to 77 years (mean 50.1 years). The urine samples of the patients in the experimental group and the control group and the urine samples of 19 normal persons (normal group) were collected. Real-timeRT-PCR method was used to detect the expression of OPN mRNA in renal tissues. Western blot method was used to detect the expression of OPN protein in renal tissues. Elisa was used to detect the concentration of OPN in urine of the three groups. The statistical data were expressed by x 鹵s and processed by SPSS14.0 software. Two groups of data were compared and analyzed by t test of two independent samples, and three groups of data were compared by single factor analysis of variance. Results: The relative expression of OPN mRNA in the experimental group and the control group was 2.04 鹵1.20 鹵2.59 鹵0.91respectively, and the relative expression of P0.05 was 0.776 鹵0.071 + 0.449 鹵0.085, respectively (P 0.05), the difference between the experimental group and the control group was significant (P 0.05), the difference between the experimental group and the control group was significant (P < 0.05). The concentration of OPN protein in urine of the normal group was 1.052 鹵0.229 鹵0.256 鹵1.498 鹵0.155, respectively. There was significant difference between the experimental group and the control group and between the experimental group and the normal group (P 0.05), but there was no significant difference between the control group and the normal group (P 0.05). Conclusion: The overexpression of OPN mRNA and protein in renal tubular epithelial cells and the decrease of OPN concentration in urine may be important factors in the formation of renal calculi in ICS patients.
