芡實對糖尿病腎病大鼠腎組織GLUT1及TGFβ1表達的影響

【摘要】:目的:探討芡實對糖尿病腎病大鼠腎組織GLUT1及TGFβ1表達的影響。 方法:60只健康雄性SD大鼠隨機分為正常對照組(N組)、糖尿病腎病組(DN)、芡實小劑量組(EL組)、芡實中劑量組(EM組)、芡實大劑量組(EH組)及氯沙坦鉀組(LP組)。采用一次性腹腔注射鏈脲佐菌素(STZ,45mg/kg)制作DN模型,造模成功后,給予EL組大鼠芡實1.5g/kg/d,EM組大鼠芡實3.0g/kg/d,EH組大鼠芡實6.0g/kg/d,,LP組大鼠氯沙坦鉀30mg/kg/d,N組及DN組給予等量蒸餾水,均采用灌胃給藥。連續(xù)灌胃12周后檢測大鼠血糖、肌酐、尿素氮、24小時尿蛋白定量;HE、Masson、PAS染色及電鏡觀察各組大鼠腎臟病理變化;免疫組化、western-blot法測定大鼠腎組織GLUT1和TGFβ1在表達情況。 結(jié)果:1.生化指標結(jié)果:實驗12周末,與正常組比較,血糖、BUN、Scr、24h尿蛋白定量顯著升高(P0.05);與DN組比較,芡實中、高劑量組大鼠病理改變減輕,BUN、Scr、24h尿蛋白定量顯著下降(P0.05);芡實中、高劑量組及氯沙坦鉀組降尿蛋白作用無統(tǒng)計學差異。2.病理改變:與正常組比較,DN組大鼠可見腎小球肥大,系膜基質(zhì)增生,基底膜增厚,芡實中、高劑量組及氯沙坦鉀組大鼠上述病理改變較DN組減輕。3.腎組織GLUT1的表達:免疫組化結(jié)果顯示,正常組大鼠GLUT1在腎小球系膜區(qū)和腎小管都有少量表達,DN組表達增強,以系膜區(qū)增強為主;與正常組比較,DN組GLUT1平均光密度值(IOD)顯著升高(P0.05),與DN組比較,芡實中、高劑量組及氯沙坦鉀組GLUT1的IOD明顯降低(P0.05);western-blot結(jié)果顯示,與正常組比較,DN組GLUT1表達明顯增強(P0.05),與DN組比較,芡實中、高劑量組及氯沙坦鉀組GLUT1表達明顯減弱(P0.05);GLUT1在免疫組化、western-blot兩種檢測方法中表達一致。4.腎組織TGFβ1的表達:免疫組化結(jié)果顯示,正常組大鼠TGFβ1在腎小球和腎小管上皮細胞都有少量表達,DN組表達增強,且以腎小管表達增強尤為顯著;與正常組比較,DN組TGFβ1的IOD值顯著升高(P0.05),與DN組比較,芡實中、高劑量組及氯沙坦鉀組TGFβ1的IOD值明顯降低(P0.05);western-blot結(jié)果顯示,與正常組比較,DN組TGFβ1表達明顯增強(P0.05),與DN組比較,芡實中、高劑量組及氯沙坦鉀組TGFβ1表達明顯減弱(P0.05);TGFβ1在免疫組化、western-blot兩種檢測方法中表達一致。 結(jié)論:芡實可能通過下調(diào)GLUT1及TGFβ1在糖尿病腎病大鼠腎組織的表達,從而延緩糖尿病腎病的進展。
[Abstract]:Aim: to investigate the effect of Euryale cornifera on the expression of GLUT1 and TGF 尾 1 in renal tissue of diabetic nephropathy rats. Methods Sixty healthy male Sprague-Dawley rats were randomly divided into two groups: normal control group (n group), diabetic nephropathy group (group D), small dose group of Euryale thicken seed group (group EL), medium dose group of Euryale seed group (group EM), large dose group of Euryale seed group (group EH) and group of Losartan potassium group (group LP). The DN model was established by a single intraperitoneal injection of STZ (45 mg / kg). After the model was successfully established, the rats in the El group were given the same amount of distilled water. The rats in the EH group were fed with the same amount of distilled water at 3.0 g / kg 路kg ~ (-1) 路d ~ (th) of Loxartan potassium, 30 mg / kg 路kg ~ (-1) D ~ (-1) N and 30 mg / kg 路kg ~ (-1) 路d ~ (-1) N of losartan, respectively. Blood glucose, creatinine, urea nitrogen in 24 hours urine protein of rats were detected by intragastric perfusion for 12 weeks, and the renal pathological changes of each group were observed by electron microscope, and the expression of GLUT1 and TGF 尾 1 in renal tissue were detected by Western blot. The result is 1: 1. Results: at the end of the 12th week of the experiment, compared with the normal group, the quantity of urine protein in 24 hours of blood glucose was significantly increased, compared with DN group, the pathological changes of the high dose group decreased significantly (P0.05), the quantity of urine protein decreased significantly in the high dose group, and in the cornale seed group, the quantity of urine protein decreased significantly in the high dose group, and in the control group, there was no significant difference between the control group and the control group. There was no significant difference between high dose group and losartan potassium group. Pathological changes: compared with normal group, glomerular hypertrophy, Mesangial matrix hyperplasia, basement membrane thickening were observed in DN group. The expression of GLUT1 in renal tissue: immunohistochemical results showed that the expression of GLUT1 in glomerular Mesangial area and renal tubule of normal rats was increased slightly in DN group, mainly in Mesangial area. Compared with the normal group, the average optical density of GLUT1 in the DN group was significantly higher than that in the DN group. Compared with the DN group, the IOD of GLUT1 in the high dose group and the losartan potassium group decreased significantly compared with the normal group. The results showed that the GLUT1 expression in the DN group was significantly higher than that in the normal group, and that in the DN group was significantly higher than that in the DN group. The expression of GLUT1 in high dose group and losartan potassium group was significantly decreased in the two methods of immunohistochemical staining, I. E. the expression of GLUT1 was consistent with that of Losartan potassium group. Expression of TGF 尾 1 in renal tissue: immunohistochemical results showed that the expression of TGF 尾 1 in glomerular and tubular epithelial cells of normal rats was increased slightly in DN group, especially in renal tubules. Compared with the normal group, the IOD value of TGF 尾 1 in the DN group was significantly higher than that in the DN group. Compared with the DN group, the IOD value of TGF 尾 1 in the medium, high dose group and losartan potassium group decreased significantly. The results showed that the expression of TGF 尾 1 in the DN group was significantly higher than that in the normal group, and compared with that in the DN group, the expression of TGF 尾 1 in the cornstarch was higher than that in the DN group. The expression of TGF 尾 1 in high dose group and losartan potassium group was significantly decreased in both immunohistochemical and Western blot methods. Conclusion: Euryale may delay the progression of diabetic nephropathy by down-regulating the expression of GLUT1 and TGF 尾 1 in renal tissue of diabetic nephropathy rats.
