全反式維甲酸對(duì)高糖誘導(dǎo)的HK-2細(xì)胞外基質(zhì)合成的影響及可能機(jī)制

【摘要】:目的探討全反式維甲酸(ATRA)對(duì)高糖誘導(dǎo)HK-2細(xì)胞外基質(zhì)合成的影響及其在延緩糖尿病腎病腎間質(zhì)纖維化中的可能作用機(jī)制。方法將體外培養(yǎng)的HK-2細(xì)胞隨機(jī)分為七組:A組(5.5 mmol/L D-葡萄糖)、B組(30 mmol/L D-葡萄糖)、C組(5.5 mmol/L D-葡萄糖+24.5 mmol/L甘露醇)、D組(10~(-7)mol/L ATRA+30 mmol/L高糖)、E組(10~(-6)mol/L ATRA+30 mmol/L高糖)、F組(10~(-5)mol/L ATRA+30 mmol/L高糖)、G組(10μmol/L Y-27632+30 mmol/L高糖),各組細(xì)胞均培養(yǎng)48 h。應(yīng)用ELISA法檢測(cè)細(xì)胞培養(yǎng)液結(jié)締組織生長(zhǎng)因子(CTGF)和Ⅳ型膠原的表達(dá)水平;RT-PCR法檢測(cè)細(xì)胞Rho A、ROCK1的表達(dá)水平。結(jié)果 B組Rho A mRNA及ROCK1 mRNA的表達(dá)較A組顯著升高(P0.05),D、E、F組Rho A mRNA及ROCK1 mRNA較B組均表達(dá)減少(P0.05),且隨著ATRA濃度的升高呈現(xiàn)劑量依賴性。G組Rho A mRNA的表達(dá)與B組無(wú)明顯差異,但ROCK1 mRNA的表達(dá)較B組顯著減少(P0.05);直線相關(guān)分析顯示高糖組,D、E、F組Rho A mRNA與ROCK1 mRNA表達(dá)呈正相關(guān)(r=0.854,P=0.030;r=0.895,P=0.016;r=0.883,P=0.020;r=0.867,P=0.025);ELISA結(jié)果顯示D、E、F組及G組CTGF、Ⅳ型膠原表達(dá)較B組顯著下降(P0.05),且下降程度與ATRA濃度呈現(xiàn)劑量相關(guān)性。結(jié)論 ATRA在一定濃度范圍內(nèi)可抑制高糖誘導(dǎo)的HK-2細(xì)胞外基質(zhì)的合成,延緩腎間質(zhì)纖維化,其機(jī)制可能與抑制Rho A/ROCK通路有關(guān)。
[Abstract]:Objective to investigate the effect of ATRAA on the synthesis of extracellular matrix of HK-2 induced by high glucose and its possible mechanism in delaying renal interstitial fibrosis of diabetic nephropathy.
