MicroRNA-21在單側(cè)輸尿管梗阻大鼠間質(zhì)纖維化腎臟組織中的表達(dá)及意義

【摘要】:目的觀察MicroRNA-21(miRNA-21)在單側(cè)輸尿管梗阻(UUO)大鼠腎臟組織中的表達(dá),初步探討其在轉(zhuǎn)化生長(zhǎng)因子-β1(TGF-β1)/Smad3致大鼠腎間質(zhì)纖維化(RIF)信號(hào)通路中的機(jī)制。方法選擇20只SD雄性大鼠,分為UUO組和假手術(shù)組(Sham組),分別于建模后第3天、第7天各采集每組5只大鼠腎臟組織,應(yīng)用實(shí)時(shí)熒光定量PCR(Real-time qPCR)方法檢測(cè)腎臟組織中miRNA-21的表達(dá)變化,采用Masson染色、蘇木素-伊紅(HE)染色和免疫組織化學(xué)技術(shù)評(píng)價(jià)腎組織纖維化,使用免疫組織化學(xué)檢測(cè)腎臟組織中TGF-β1、Smad3、Ⅰ型膠原蛋白(Col-Ⅰ)和α平滑肌肌動(dòng)蛋白(α-SMA)表達(dá)情況。結(jié)果建模后第3天和第7天UUO組腎臟組織中miRNA-21與Sham組比較均升高(P0.01),7dUUO組腎組織中miRNA-21與3dUUO組相比升高(P0.01)。建模后3、7dUUO組Masson染色纖維化評(píng)分、TGF-β1、Samd3、α-SMA及Col-Ⅰ蛋白陽性表達(dá)面積較Sham組升高(P0.01),以上指標(biāo)7dUUO組較3dUUO組比較表達(dá)增強(qiáng)(P0.01)。UUO組腎組織中miRNA-21的表達(dá)與腎間質(zhì)纖維化評(píng)分正相關(guān)(r=0.888,P0.01);腎組織miRNA-21與TGF-β1、Smad3、Col-Ⅰ、α-SMA蛋白表達(dá)呈正相關(guān)(r=0.799、0.849、0.882、0.896,P0.01)。結(jié)論 UUO大鼠腎組織中miRNA-21在建模后表達(dá)上調(diào),且間質(zhì)纖維化程度越重表達(dá)越高,TGF-β1/Smad3信號(hào)通路可能是通過上調(diào)miRNA-21從而介導(dǎo)大鼠腎間質(zhì)纖維化。
[Abstract]:Objective to investigate the expression of microRNA-21 miRNA-21 in the renal tissue of rats with unilateral ureteral obstruction (UUO), and to explore the mechanism of microRNA-21 miRNA-21 in the signal pathway of TGF- 尾 1)/Smad3 induced by TGF- 尾 1)/Smad3 in renal interstitial fibrosis of rats with unilateral ureteral obstruction (UUO). Methods Twenty Sprague-Dawley male rats were divided into UUO group and sham-operated group. The kidney tissues of each group were collected on the 3rd and 7th day after modeling.
