PSCA基因多態(tài)性與膀胱癌易感性關(guān)系的Meta分析

【摘要】:目的:進(jìn)一步探討前列腺干細(xì)胞抗原(PSCA)基因rs2294008多態(tài)性與膀胱癌易感性的關(guān)系。方法:系統(tǒng)檢索PubMed、MEDLINE、EMBASE、Cochrane圖書館、中國生物醫(yī)學(xué)數(shù)據(jù)庫(CBM)、相關(guān)期刊論文(CNKI)及萬方數(shù)據(jù)庫中2016年10月之前的有關(guān)PSCA基因rs2294008多態(tài)性與膀胱癌易感性相關(guān)的研究。嚴(yán)格制定納入及排除標(biāo)準(zhǔn),提取數(shù)據(jù),采用NOS量表進(jìn)行質(zhì)量評(píng)價(jià),應(yīng)用Stata12.0軟件進(jìn)行Meta分析,應(yīng)用OR值及其95%CI評(píng)估其關(guān)聯(lián)強(qiáng)度,并進(jìn)行發(fā)表偏倚評(píng)估分析。結(jié)果:共納入文獻(xiàn)8篇,包括13個(gè)病例-對(duì)照研究,涉及12349病例及24836例對(duì)照。Meta分析結(jié)果顯示:在5種基因模型中PSCA rs2294008(CT)多態(tài)性與膀胱癌易感性均顯著相關(guān),進(jìn)一步按種族及病例來源進(jìn)行亞組分析,等位基因頻率模型(T vs C)[OR=1.151,95%CI=1.12-1.19,P=0.0000.001,I~2=0.0%];顯性模型(CT+TT vs CC)=[OR=1.271,95%CI=1.20-1.33,P=0.0000.001,I~2=0.0%]、隱性模型(TT vs CC+CT)=[OR=1.151,95%CI=1.08-1.21,P=0.0000.001,I~2=0.0%]、純合子模型(TT vs CC)=[OR=1.311,95%CI=1.22-1.40,P=0.0000.001,I~2=0.0%]及共顯性模型(CT vs CC)=[OR=1.25,95%CI=1.18-1.32,P=0.0000.001,I~2=0.0%],以上模型分析結(jié)果均說明:PSCA rs2294008(CT)多態(tài)性與膀胱癌顯著相關(guān)。結(jié)論:PSCA rs2294008(CT)基因多態(tài)性與膀胱癌的易感性顯著相關(guān),且與種族及病例來源無關(guān);突變型T等位基因可能為膀胱癌的易感基因位點(diǎn),PSCA有望成為膀胱癌遺傳易感性的生物學(xué)標(biāo)記物。
[Abstract]:Objective: to investigate the relationship between rs2294008 polymorphism of prostate stem cell antigen (PSCA) gene and bladder cancer susceptibility.Methods: a systematic search was conducted on the PSCA gene rs2294008 polymorphism and its association with bladder cancer susceptibility in PubMedus MEDLINE Cochrane Library, Chinese Biomedical Database (CBM), Chinese Journal Full-text Database (CNKI) and Wanfang database before October 2016.The inclusion and exclusion criteria were strictly formulated, the data were extracted, the quality was evaluated by NOS scale, the Meta was analyzed by Stata12.0 software, the correlation strength was evaluated by OR value and its 95%CI, and the evaluation of publication bias was carried out.Results: a total of 8 articles were included, including 13 case-control studies, involving 12349 cases and 24836 controls.
