循環(huán)miR-152對前列腺癌生化復(fù)發(fā)早期預(yù)測的研究

【摘要】:目的:探討循環(huán)微小RNA 152(miR-152)對前列腺癌生化復(fù)發(fā)早期預(yù)測的價(jià)值。方法:收集將要進(jìn)行前列腺癌手術(shù)的患者66例(生化復(fù)發(fā)組:35例,術(shù)后2年內(nèi)生化復(fù)發(fā);未生化復(fù)發(fā)組:31例,術(shù)后2年內(nèi)未生化復(fù)發(fā)),另擇年齡相匹配的31例健康男性作為健康對照組。用qRT-PCR法檢測前列腺癌患者和健康對照組的循環(huán)miR-152表達(dá)水平。應(yīng)用ROC曲線分析循環(huán)miR-152對前列腺癌早期生化復(fù)發(fā)的診斷價(jià)值。分析復(fù)發(fā)組循環(huán)miR-152表達(dá)水平與臨床病理參數(shù)(年齡、術(shù)前PSA水平、Gleason積分、臨床分期、骨轉(zhuǎn)移)之間的相關(guān)性。結(jié)果:循環(huán)miR-152在前列腺癌術(shù)后患者(未生化復(fù)發(fā)及生化復(fù)發(fā))與健康對照組的表達(dá)差異有統(tǒng)計(jì)學(xué)意義(t=-5.212,P=0.001)。前列腺癌術(shù)后生化復(fù)發(fā)組患者循環(huán)miR-152的表達(dá)量較未生化復(fù)發(fā)組患者低,差異有統(tǒng)計(jì)學(xué)意義(t=-5.727,P=0.001)。循環(huán)miR-152對前列腺癌術(shù)后生化復(fù)發(fā)早期預(yù)測的ROC曲線下面積(AUC)為0.906(95%CI,0.809~0.964),靈敏度和特異度分別為91.4%、80.6%。循環(huán)miR-152表達(dá)水平與Gleason積分、臨床分期、有無生化復(fù)發(fā)和骨轉(zhuǎn)移相關(guān)(P均0.05);隨著Gleason積分、臨床分期的增加,循環(huán)miR-152的表達(dá)水平下降。但miR-152表達(dá)量與患者年齡、術(shù)前PSA無關(guān)(P均0.05)。結(jié)論:循環(huán)miR-152表達(dá)水平在前列腺癌術(shù)后生化復(fù)發(fā)患者中顯著降低,可能成為前列腺癌生化復(fù)發(fā)早期預(yù)測的分子標(biāo)志物。
[Abstract]:Objective: to investigate the value of circulating minimal RNA 152miR-152 in early prediction of biochemical recurrence of prostate cancer. Methods: 66 patients (35 cases of biochemical recurrence group, 35 cases of biochemical recurrence group and 31 cases of non-biochemical recurrence group) were collected. The circulating miR-152 expression in prostate cancer patients and healthy control group was detected by qRT-PCR method. The circulating miR-152 expression was analyzed by ROC curve. Diagnostic value of early biochemical recurrence of prostate cancer: analysis of circulating miR-152 expression and clinicopathologic parameters (age, age). Preoperative PSA level, Gleason score, clinical stage, Results: the expression of circulating miR-152 in postoperative patients with prostate cancer was significantly different from that in healthy control group. The expression of cyclic miR-152 was lower than that of non-biochemically relapsed patients. The area under the ROC curve of circulating miR-152 for the early prediction of postoperative biochemical recurrence of prostate cancer was 0.906 ~ 95CII 0.809 ~ 0.964U, the sensitivity and specificity were 91.4 ~ 80.6.The circulating miR-152 expression level, Gleason integral, clinical stage, and so on. The expression of circulating miR-152 decreased with the increase of Gleason score and clinical stage, but the expression of miR-152 was correlated with the age of the patients. Conclusion: the expression of circulating miR-152 was significantly decreased in patients with postoperative recurrence of prostate cancer and might be a molecular marker for early prediction of biochemical recurrence of prostate cancer.
