高糖環(huán)境下纈沙坦對腎小管上皮細胞-間充質轉化過程中骨膜蛋白、轉錄激活因子1的影響

【摘要】:目的:研究在高糖環(huán)境下纈沙坦對腎小管上皮細胞-間充質轉化(EMT)過程中骨膜蛋白(Periostin)、轉錄激活因子1(AP-1)的影響。方法:體外高糖環(huán)境下培養(yǎng)人腎小管上皮細胞(HK-2細胞),分為空白對照組和纈沙坦高、中、低劑量組(10~(-3)、10~(-4)、10~(-5)mol/L),作用48 h;另設正常對照組。觀察各組HK-2細胞形態(tài)學變化,檢測各組HK-2細胞活力(OD值),細胞中Ⅲ型膠原含量、AP-1蛋白表達和Periostin、AP-1 mRNA表達。結果:與正常對照組比較,其余各組HK-2細胞部分細胞間隙增大并與附近的細胞脫離接觸,細胞分裂旺盛,細胞活力、Ⅲ型膠原含量、AP-1蛋白表達和Periostin、AP-1 mRNA表達均增強(P0.05)。與空白對照組比較,纈沙坦高、中、低劑量組細胞活力、Ⅲ型膠原含量、AP-1蛋白表達和Periostin、AP-1 mRNA表達均更低(P0.05),其中纈沙坦中劑量組作用最明顯(P0.01)。結論:體外高糖環(huán)境下可促進HK-2細胞EMT過程;纈沙坦可通過抑制AP-1的過表達進而下調轉分化的腎小管上皮細胞中Periostin蛋白表達,從而減緩EMT過程。
[Abstract]:Objective: to study the effect of valsartan on the expression of periostein-Periostin1 (transcriptional activator 1) in the process of renal tubular epithelial cell-mesenchymal transformation (EMTT). Methods: human renal tubular epithelial cells (HK-2) were cultured in high glucose environment in vitro. Cystonia, divided into blank control group and valsartan high, In the low dose group, the cells of HK-2 were observed in morphologic changes, OD value of HK-2 cells, and the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells. Results: compared with the normal control group, the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells were compared with those in the normal control group, and the morphological changes of the HK-2 cells in each group were observed, and the OD value of HK-2 cells in each group was measured, and the expression of AP-1 protein and PeriostinAP-1 mRNA in the cells were compared with those in the normal control group. In other groups, the gap of some HK-2 cells was enlarged and disconnected from the nearby cells.
