水飛薊賓對人膀胱癌細(xì)胞T24和5637的增殖抑制及凋亡誘導(dǎo)作用

【摘要】:目的:探討水飛薊賓(SB)對人膀胱癌細(xì)胞T24及5637增值及凋亡的影響,初步闡明其可能的機(jī)制。 方法:培養(yǎng)人膀胱癌細(xì)胞5637和T24,取處于對數(shù)生長期的5637和T24細(xì)胞,,分別設(shè)細(xì)胞對照組及25、50、100、200和400μmol·L-1SB實(shí)驗(yàn)組,MTT法檢測SB對人膀胱癌細(xì)胞系T24和5637的增殖抑制作用;細(xì)胞劃痕實(shí)驗(yàn)在倒置顯微鏡下觀察不同濃度SB作用不同時(shí)間對人膀胱癌細(xì)胞系T245637的侵襲抑制能力;DAPI染色法觀察給藥后細(xì)胞形態(tài)的變化;流式細(xì)胞術(shù)檢測并評估SB誘導(dǎo)腫瘤細(xì)胞凋亡的能力。 結(jié)果:MTT檢測隨著SB劑量的增加和作用時(shí)間延長,不同濃度SB組T24和5637細(xì)胞存活率顯著低于對照組(p<0.05);不同劑量SB處理后,各組細(xì)胞的遷移明顯受到抑制;DAPI染色及流式細(xì)胞術(shù)檢測,不同劑量SB均能夠有效誘導(dǎo)T24和5637細(xì)胞發(fā)生凋亡,凋亡率顯著高于對照組(p<0.05)。 結(jié)論:SB通過誘導(dǎo)人膀胱癌細(xì)胞系T24和5637細(xì)胞凋亡從而抑制細(xì)胞侵襲及增殖。
[Abstract]:Objective: To investigate the effect of silybin (SB) on the increment and apoptosis of human bladder cancer cell line T24 and 5637, and to elucidate its possible mechanism.
Methods: cultured human bladder cancer cell T24 5637 and 5637, in the logarithmic growth phase and T24 cells, respectively. Cells in the control group and 25,50100200 and 400 mol L-1SB experimental group, inhibition of SB MTT detection method on human bladder cancer cell line T24 and 5637 proliferation; on human bladder cancer cell line T245637 the effects of different concentrations of SB inhibit the invasion ability of different time under inverted microscope, cell scratch assay; DAPI staining was used to observe the cell morphology changes after administration; the ability of flow cytometry to detect and evaluate the SB induced apoptosis in tumor cells.
Results: the detection of MTT with the increase of SB dose and time prolonging, different concentration of SB groups and T24 5637 cell survival rate was significantly lower than the control group (P < 0.05); different doses of SB after treatment, cell migration were significantly inhibited; DAPI staining and flow cytometry, different doses of SB can effectively 5637 and T24 induced cell apoptosis, the apoptosis rate was significantly higher than the control group (P < 0.05).
Conclusion: SB inhibits cell invasion and proliferation by inducing apoptosis of human bladder cancer cell line T24 and 5637 cells.
