miRNA-196a2及miRNA-499基因多態(tài)性與膀胱癌的相關(guān)性研究

【摘要】:目的微小RNA(microRNA,miRNA)基因多態(tài)性與腫瘤的關(guān)系密切。文中探討rs11614913(miRNA-196a2)及rs3746444(miRNA-499)的基因多態(tài)性與膀胱癌及其病理參數(shù)的相關(guān)性。方法選取膀胱癌患者283人作為膀胱癌組,正常體檢者283人作為正常對(duì)照組。利用Massarray單核苷酸多態(tài)性檢測(cè)技術(shù)對(duì)膀胱癌組和正常對(duì)照組的rs11614913(miRNA-196a2)及rs3746444(miRNA-499)基因多態(tài)性進(jìn)行分析;采用Logistic回歸模型分析各基因型與膀胱癌發(fā)生的關(guān)系并比較不同基因型與膀胱癌及其病理參數(shù)的關(guān)系。結(jié)果 rs11614913位點(diǎn)的各基因型在2組中分布差異有統(tǒng)計(jì)學(xué)意義(χ2=15.077,P0.001),其中TT基因型在膀胱癌組中的頻率(19.08%)顯著低于對(duì)照組(33.95%),差異有統(tǒng)計(jì)學(xué)意義(P0.05);TT基因型發(fā)生膀胱癌的風(fēng)險(xiǎn)是CC基因型的0.45倍(ORadjusted=0.45,95%CI:0.28~0.73)。rs3746444位點(diǎn)的各基因型在2組中分布差異無(wú)統(tǒng)計(jì)學(xué)意義(χ2=5.107,P=0.078),但GG基因型在膀胱癌組中分布頻率(7.77%)顯著高于對(duì)照組(3.56%),差異有統(tǒng)計(jì)學(xué)意義(P0.05);GG基因型發(fā)生膀胱癌的風(fēng)險(xiǎn)是AA的2.39倍(ORadjusted=2.39,95%CI:1.10~5.17)。rs11614913及rs3746444位點(diǎn)的基因型分布頻數(shù)在不同腫瘤分期、周圍淋巴結(jié)轉(zhuǎn)移、遠(yuǎn)端淋巴結(jié)轉(zhuǎn)移組間的差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。然而,在不同分化程度的腫瘤分組中2個(gè)位點(diǎn)的基因型分布頻數(shù)差異有統(tǒng)計(jì)學(xué)意義(rs11614913:χ2=15.066,P=0.005;rs3746444:χ2=11.120,P=0.025)。結(jié)論 rs11614913(miRNA-196a2)及rs3746444(miRNA-499)基因多態(tài)性與膀胱癌的發(fā)生相關(guān),2個(gè)位點(diǎn)的基因多態(tài)性與腫瘤的分化程度相關(guān)。
[Abstract]:Objective to investigate the association between microRNA-miRNA-499) gene polymorphisms and bladder cancer and its pathological parameters. Methods 283 patients with bladder cancer were selected as bladder cancer group, and the relationship between the polymorphism of rs11614913 miRNA-196a2) and rs3746444 miRNA-499) gene polymorphism was studied. The rs11614913 miRNA-196a2 and rs3746444miRNA-499) gene polymorphisms were detected by Massarray single nucleotide polymorphisms (SNP) in normal controls. Logistic regression model was used to analyze the relationship between genotypes and bladder carcinogenesis, and to compare the relationship between different genotypes and bladder cancer and its pathological parameters. Results there were significant differences in the distribution of rs11614913 loci between the two groups (蠂 2 = 15.07 7, P 0.001). The frequency of TT genotype in bladder cancer group (19.08) was significantly lower than that in control group (33.95%). The difference was statistically significant. The risk of bladder cancer of TT genotype was 0.45 times higher than that of CC genotype. The frequency of GG genotype distribution in bladder cancer group was significantly higher than that in control group (7.77). There was a statistically significant difference in the risk of bladder cancer with P0.05 GG genotype. The risk of bladder cancer was 2.39 times higher than that of AA, and the distribution frequency of GG genotype was 2.39 times higher than that of AA, and the distribution frequency of Rs11614913 and rs3746444 locus was different in different stages of the tumor, and the frequency of GG genotype distribution was significantly higher than that of control group (P < 0.05), but the distribution frequency of GG genotype in bladder cancer group was significantly higher than that in control group (P = 3.56). The risk of GG genotype was 2.39 times higher than that of AA. There was no significant difference between the peripheral lymph node metastasis group and the distal lymph node metastasis group (P 0.05). There were significant differences in the genotypic frequencies of two loci in different differentiated tumor groups. There were significant differences in the genotypic frequencies of two loci: rs11614913: 蠂 ~ 2 ~ 2 ~ (15.0666) P ~ (0.005) rs37466 ~ (4): 蠂 ~ (2) ~ (2) ~ (11.120) (P ~ (0.02525)). Conclusion the polymorphism of rs11614913 miRNA-196a2 and rs3746444miRNA-499) gene polymorphisms are associated with the development of bladder cancer, and the gene polymorphisms of two loci are related to the differentiation degree of the tumor.
