【摘要】：目的建立適合治療給藥的大鼠抗腎小球基底膜(GBM)腎炎模型。方法采用家兔多點(diǎn)sc大鼠腎小球基底膜與弗氏佐劑乳化物的方法進(jìn)行免疫,兩周1次,共5次,制備抗血清;分次給大鼠iv抗血清,檢測(cè)血清循環(huán)免疫復(fù)合物(CIC)水平、腎小球基底膜免疫熒光強(qiáng)度、腎臟及腎小球基底膜病變,確定適合治療給藥的抗血清效價(jià)、免疫劑量、方式及時(shí)間。以固腎2號(hào)為治療藥物,驗(yàn)證模型的可行性。結(jié)果使用效價(jià)為1∶4的兔抗大鼠GBM血清4 m L,分3 d 4次iv免疫,免疫第6、8、10天,均能形成大鼠抗腎小球基底膜腎炎,免疫第6天,病變相對(duì)較輕,免疫第8天及第10天,病變基本穩(wěn)定。驗(yàn)證實(shí)驗(yàn)結(jié)果顯示,連續(xù)給藥30 d,固腎2號(hào)顯著降低模型大鼠24 h尿蛋白排出,減輕腎臟病變,對(duì)大鼠抗腎小球基底膜腎炎有明顯治療作用。結(jié)論使用效價(jià)為1∶4的兔抗大鼠GBM血清4 m L,分3 d 4次iv免疫,免疫后8 d,能形成適合治療給藥的大鼠抗腎小球基底膜腎炎模型。
[Abstract]:Objective to establish a rat model of anti-glomerular basement membrane glomerulonephritis (GBM) which is suitable for treatment. Methods the rat glomerular basement membrane was immunized with Freund's adjuvant emulsifier once a fortnight by the method of rabbit multi-point sc rat glomerular basement membrane and Freund's adjuvant emulsification. Antiserum was prepared for 5 times. The serum level of circulating immune complex (CICC), the immunofluorescence intensity of glomerular basement membrane and the pathological changes of renal and glomerular basement membrane were determined to determine the titer of antiserum suitable for treatment. The dose, mode and time of immunization. The feasibility of the model was verified by using Gushen 2 as the therapeutic drug. Results 4 ml of rabbit anti-rat GBM serum with titer of 1: 4 was used and immunized for 3 days and 4 times iv. On day 6 and day 10, all rats developed anti-glomerular basement membrane glomerulonephritis. On the 6th day of immunization, the lesion was relatively mild, and on the 8th and 10th day of immunization, the lesion was basically stable. The experimental results showed that the lesion was basically stable. After 30 days of continuous administration, Gushen No. 2 significantly decreased the excretion of urinary protein and alleviated the renal lesion in the model rats. Conclusion Rabbit anti-rat GBM serum with titer of 1: 4 was used for 4 mL, 3 days and 4 times iv immunization, 8 days after immunization. The model of anti-glomerular basement membrane glomerulonephritis was established in rats suitable for treatment.
