小鼠不同部位體內(nèi)前列腺癌骨轉(zhuǎn)移模型的建立及方法改進

【摘要】:目的:構(gòu)建并改進前列腺癌(prostate cancer,PCa)體內(nèi)股骨及脊柱轉(zhuǎn)移動物模型,為不同部位的PCa骨轉(zhuǎn)移的診治及骨應(yīng)力改變的研究提供工具。方法:將不同濃度的PCa細(xì)胞(RM-1)注射到小鼠(C57BL/6)股骨骨髓腔和腰椎內(nèi),觀察記錄小鼠活動變化、成瘤情況,測定瘤體大小及記錄存活時間。取模型的雙側(cè)股骨組織,進行影像學(xué)檢查后測算灰度值(骨密度),并行組織病理學(xué)檢查,確定建模情況及成瘤類型。結(jié)果:各細(xì)胞濃度梯度小鼠均成瘤,各股骨轉(zhuǎn)移模型的腫瘤發(fā)生時間、生長速度同組間無差別(P0.05),病理學(xué)檢測結(jié)果證實所有股骨骨髓腔和腰椎內(nèi)PCa骨轉(zhuǎn)移模型構(gòu)建成功,且較低RM-1細(xì)胞濃度構(gòu)建的PCa骨轉(zhuǎn)移動物模型生存時間較前期構(gòu)建的動物模型明顯延長(約1倍,至少2周)。結(jié)論:成功用同一PCa細(xì)胞株構(gòu)建了不同部位(四肢骨和中軸骨)的PCa骨轉(zhuǎn)移體內(nèi)動物模型,經(jīng)股骨骨髓腔穿刺注射腫瘤細(xì)胞建立PCa骨轉(zhuǎn)移動物模型成瘤率高,腫瘤生長迅速,且可穩(wěn)定復(fù)制,為進一步PCa骨轉(zhuǎn)移的骨應(yīng)力改變及防治的研究提供平臺。
[Abstract]:Objective: to establish and improve the animal model of femoral and spinal metastasis in prostate cancer. To provide a tool for the diagnosis and treatment of PCa bone metastasis and the study of bone stress changes in different sites. Methods: different concentrations of PCa cells were injected into C57BL / 6 mice. Femoral medullary cavity and lumbar vertebrae. The changes of activity, tumorigenesis, tumor size and survival time were observed and recorded. The bilateral femur tissues of the model were taken and the gray values (bone density and histopathology) were measured after imaging examination. Results: each cell concentration gradient mice were tumorigenic, and the tumorigenesis time and growth rate of each femur metastasis model were not different (P0.05). Pathological findings confirmed that all femoral medullary and lumbar PCa bone metastasis models were successfully constructed. The survival time of PCa bone metastasis animal model with low RM-1 cell concentration was significantly longer than that of the previous model (about twice as long). Conclusion: the animal model of PCa bone metastasis in different parts (limb bone and midaxial bone) was successfully constructed with the same PCa cell line. The animal model of PCa bone metastasis was established by injecting tumor cells through femoral medullary cavity with high tumorigenesis rate, rapid tumor growth and stable replication. To provide a platform for further study of bone stress change and prevention and treatment of PCa bone metastasis.
