鎂離子對高磷誘導(dǎo)的大鼠胸主動脈血管環(huán)鈣化的影響

【摘要】:目的探討不同濃度鎂離子對高磷誘導(dǎo)的大鼠胸主動脈血管環(huán)鈣化的影響。方法體外分離大鼠胸主動脈血管環(huán),將其隨機分為正常對照組(含10%胎牛血清DMEM培養(yǎng)基)、高磷組(含10 mmol/Lβ-甘油磷酸高磷培養(yǎng)基)和鎂干預(yù)組(高磷培養(yǎng)基加不同濃度硫酸鎂,使鎂離子終濃度分別為1、2、3 mmol/L,鎂干預(yù)組Ⅰ~Ⅲ)。給予14天干預(yù)后,采用von Kossa染色及鄰甲酚酞絡(luò)合酮比色法檢測胸主動脈血管環(huán)鈣化情況;免疫組織化學(xué)方法檢測胸主動脈血管環(huán)L型鈣通道α1c(LTCCα1c)、LTCCβ3亞基、Runt相關(guān)轉(zhuǎn)錄因子2(Runx2)和平滑肌22α(SM22α)表達情況。結(jié)果與正常對照組比較,高磷組鈣沉積增加,鎂干預(yù)組隨鎂離子濃度增大棕黑色顆粒沉積逐漸減少,鈣含量測定結(jié)果與之相一致(P0.05)。Runx2、LTCCα1c、LTCCβ3亞基的表達除鎂干預(yù)組Ⅲ與正常對照組差異無統(tǒng)計學(xué)意義外,其余各組均高于正常對照組(P0.05);鎂干預(yù)組隨鎂離子濃度增大,Runx2、LTCCα1c、LTCCβ3亞基的表達水平均逐漸降低,且均低于高磷組(P0.05)。SM22α表達除鎂干預(yù)組Ⅲ與正常對照組差異無統(tǒng)計學(xué)意義外,其余各組均低于正常對照組(P0.05)。相關(guān)性分析顯示,LTCCβ3、LTCCα1c亞基均與Runx2表達呈正相關(guān)(r=0.692,P0.001;r=0.716,P0.001),Runx2與SM22α表達呈負相關(guān)(r=-0.671,P=0.001)。結(jié)論鎂離子在一定程度上可以抑制高磷誘導(dǎo)的大鼠胸主動脈血管環(huán)鈣化,其可能通過下調(diào)LTCCα1c、LTCCβ3亞基表達,抑制VSMCs向成骨/成軟骨表型轉(zhuǎn)化,進而抑制血管環(huán)鈣化的發(fā)生。
[Abstract]:Objective to investigate the effects of different concentrations of magnesium ions on calcification of thoracic aortic rings induced by high phosphorus in rats. They were randomly divided into normal control group (containing 10% fetal bovine serum DMEM medium). The final concentration of magnesium ion in high phosphorus group (containing 10 mmol/L 尾 -glycerophosphoric acid medium) and magnesium intervention group (high phosphorus medium plus different concentration of magnesium sulfate) was 1 ~ 2 ~ 2 ~ 3 mmol/L, respectively. After 14 days of intervention, calcification of thoracic aortic rings was detected by von Kossa staining and o-cresol phthalein complexone colorimetry. The L-type calcium channel 偽 1cTCC 偽 1cc and LTCC 尾 3 subunit of thoracic aortic rings were detected by immunohistochemical method. The expression of Runt related transcription factor 2nRunx2) and smooth muscle 22 偽 -SM22 偽. Results compared with the normal control group, calcium deposition increased in the high phosphorus group. In the magnesium intervention group, the deposition of brown black granules decreased with the increase of magnesium ion concentration, and the results of calcium content were consistent with that of P0.05N.
