研究背景:哮喘是由于遺傳和環(huán)境因素所導(dǎo)致的一種復(fù)雜的慢性氣道疾病,遺傳度為30-70%。隨著全基因組關(guān)聯(lián)分析方法大規(guī)模的應(yīng)用,越來越多的哮喘易感基因被鑒定,但由于人種的差異,這些結(jié)果需要在不同的人群中進行重復(fù)和驗證。2006年,一個德國研究小組利用定位候選克隆方法在5p13區(qū)域發(fā)現(xiàn)了包括IL7R在內(nèi)的多個哮喘易感位點。由于該區(qū)域在多個人群中已反復(fù)被證實是一個與哮喘連鎖的一個區(qū)域,而且在這個區(qū)域內(nèi)含有多個細(xì)胞因子相關(guān)的基因。鑒于以上證據(jù),加之考慮到IL7R的功能,我們認(rèn)為IL7R是一個哮喘的候選基因。 目的:本研究的主要目的是利用基于群體的病例對照關(guān)聯(lián)分析方法探討白細(xì)胞介素7受體基因在漢族人群哮喘發(fā)生中的作用。 材料和方法:收集哮喘患者及正常對照血樣并提取基因組DNA,所收集的400個哮喘患者均符合支氣管哮喘診斷標(biāo)準(zhǔn),424個正常對照均無過敏性哮喘及其他過敏性疾病的癥狀及病史并排除肝、腎疾病。全部標(biāo)本來自濰坊哮喘病醫(yī)院及山東大學(xué)齊魯醫(yī)院,病例組個體與對照組個體均為年齡和性別匹配的山東漢族人。 利用Haploview分析來自HapMap數(shù)據(jù)庫中HCB人群的IL7R基因的數(shù)據(jù),我們從中選擇了...
