卵巢癌組織Hippo-Yes相關(guān)蛋白表達(dá)與臨床病理參數(shù)及增殖凋亡相關(guān)性研究

【摘要】:目的探討卵巢癌組織中Hippo-Yes相關(guān)蛋白(Yes-associated protein,YAP)的表達(dá)與臨床病理參數(shù)的關(guān)系,及其對卵巢癌細(xì)胞增殖和凋亡的影響。方法免疫組化染色技術(shù)檢測湖南省腫瘤醫(yī)院病理科2010-01-01-2013-12-31保存的68例卵巢癌及38例良性卵巢組織中YAP蛋白表達(dá),分析YAP表達(dá)與卵巢癌臨床病理參數(shù)的相關(guān)性;逆轉(zhuǎn)錄病毒介導(dǎo)的YAP shRNA干擾人卵巢癌EFO-21細(xì)胞中YAP表達(dá),蛋白質(zhì)印跡法檢測干擾效率,5-溴脫氧尿嘧啶核苷(5-Bromo-2-deoxy Uridine,BrdU)細(xì)胞增殖分析和流式細(xì)胞術(shù)檢測腫瘤細(xì)胞增殖和凋亡變化。結(jié)果卵巢癌組織YAP蛋白陽性表達(dá)率為61.8%(42/68),良性卵巢組織為10.5%(4/38),差異有統(tǒng)計(jì)學(xué)意義,χ2=26.054,P0.001。卵巢癌組織中YAP蛋白陽性表達(dá)率在高臨床分期(χ2=8.600,P=0.003)和高病理分級(χ2=5.688,P=0.017)中顯著升高。YAP shRNA顯著降低卵巢癌EFO-21細(xì)胞中YAP蛋白表達(dá),對照組YAP蛋白表達(dá)為0.96±0.16,實(shí)驗(yàn)組為0.14±0.04,P0.001。YAP shRNA導(dǎo)致腫瘤細(xì)胞增殖能力顯著減弱,對照組細(xì)胞增殖水平為121.00±3.19,實(shí)驗(yàn)組為55.83±3.56,P0.001。YAP shRNA并誘導(dǎo)細(xì)胞凋亡,對照組凋亡率為(8.75±0.15)%,實(shí)驗(yàn)組為(32.75±4.76)%,P0.001。結(jié)論 YAP蛋白在卵巢癌組織中表達(dá)異常升高并核內(nèi)蓄積,其陽性表達(dá)與高病理分級和高臨床分期顯著有關(guān),干擾YAP表達(dá)導(dǎo)致腫瘤細(xì)胞增殖減弱和凋亡增加,提示YAP通過促進(jìn)腫瘤細(xì)胞生長發(fā)揮促癌作用,其可能成為卵巢癌靶向治療的潛在靶點(diǎn)。
[Abstract]:Objective to investigate the relationship between the expression of Hippo-Yes associated protein (Yes-associated protein,YAP) and clinicopathological parameters in ovarian cancer and its effect on proliferation and apoptosis of ovarian cancer cells. Methods Immunohistochemical staining was used to detect the expression of YAP protein in 68 cases of ovarian cancer and 38 cases of benign ovarian tissue preserved in the Department of Pathology, Hunan Cancer Hospital. The correlation between the expression of YAP and the clinicopathological parameters of ovarian cancer was analyzed. Retrovirus-mediated YAP shRNA interfered with the expression of YAP in human ovarian cancer EFO-21 cells. Western imprinting was used to detect the interference efficiency. 5-bromodeoxyuridine (5-Bromo-2-deoxy Uridine,BrdU) cell proliferation analysis and flow cytometry were used to detect the proliferation and apoptosis of human ovarian cancer cells.
