低氧微環(huán)境與宮頸癌耐藥的關(guān)系

【摘要】:目的通過(guò)檢測(cè)低氧誘導(dǎo)因子-1α(HIF-1α)、多耐藥基因1(MDR1)在人宮頸癌組織、He La細(xì)胞中的表達(dá)及兩者間的關(guān)系,探討低氧微環(huán)境與宮頸癌耐藥的關(guān)系。方法應(yīng)用免疫組化法檢測(cè)78例中、晚期宮頸癌組織微陣列芯片組織標(biāo)本中HIF-1α、MDR1的表達(dá);以二氯化鈷(Co Cl2)處理He La細(xì)胞模擬細(xì)胞低氧微環(huán)境,設(shè)置低氧組、常氧組。細(xì)胞免疫熒光法檢測(cè)HIF-1α、MDR1蛋白在He La細(xì)胞中表達(dá)。結(jié)果 HIF-1α、MDR1在宮頸癌組織中的陽(yáng)性表達(dá)率分別為69.2%、85.6%,且表達(dá)呈正相關(guān)(r=0.348,P0.05)。低氧組He La細(xì)胞HIF-1α、MDR1表達(dá)分別為34.76±7.09、57.61±57.61,顯著高于常氧組的19.46±3.57、29.57±12.85(P0.05),低氧組HIF-1α與MDR1表達(dá)呈正相關(guān)(r=0.315,P0.05)。結(jié)論低氧微環(huán)境與宮頸癌耐藥發(fā)生有關(guān),可能的機(jī)制是HIF-1α促進(jìn)MDR1的表達(dá)從而導(dǎo)致耐藥的發(fā)生。
[Abstract]:Objective to detect the expression of hypoxia inducible factor-1 偽 (HIF-1 偽) and multi-resistance gene 1 (MDR1) in human cervical cancer, He La cells and their relationship, and to explore the relationship between hypoxia microenvironment and drug resistance in cervical cancer. Methods Immunohistochemical method was used to detect the expression of HIF-1 偽 and MDR1 in 78 cases of advanced cervical cancer tissue microarray tissue, and He La cells were treated with cobalt dichloride (Co Cl2) to simulate the hypoxia microenvironment of the cells, and the hypoxia group and normoxic group were set up.
