MicroRNA-18a及雌激素受體α在子宮肌瘤中的表達(dá)

【摘要】:目的檢測(cè)子宮肌瘤及瘤旁正常平滑肌組織中microRNA-18a(miR-18a)及雌激素受體α(ERα)的表達(dá)差異,探討microRNA-18a及ERα與子宮肌瘤的關(guān)系。方法用原位雜交法和免疫組化法分別檢測(cè)microRNA-18a和ERα在子宮肌瘤及瘤旁正常平滑肌組織石蠟切片中的表達(dá),并評(píng)價(jià)microRNA-18a及ERα的表達(dá)之間的關(guān)系。結(jié)果子宮肌瘤組織中microRNA-18a表達(dá)顯著低于瘤旁正常平滑肌組織(P0.05),而ERα表達(dá)顯著高于瘤旁正常平滑肌組織(P0.05)。子宮肌瘤組織及瘤旁正常平滑肌組織中microRNA-18a及ERα的表達(dá)量呈負(fù)相關(guān)。結(jié)論 microRNA-18a可能通過(guò)影響ERα在子宮肌瘤中的表達(dá),從而在子宮肌瘤發(fā)生發(fā)展過(guò)程中起到重大作用;microRNA-18a可能成為治療子宮肌瘤的新靶標(biāo)。
[Abstract]:Objective to detect the expression of microRNA-18a (miR-18a) and estrogen receptor 偽 (ER 偽) in uterine myoma and adjacent normal smooth muscle tissues, and to explore the relationship between microRNA-18a and ER 偽 and uterine myoma. Methods the expression of microRNA-18a and ER 偽 in paraffin sections of uterine myoma and normal smooth muscle tissue adjacent to tumor was detected by in situ hybridization and immunohistochemistry, respectively, and the relationship between the expression of microRNA-18a and ER 偽 was evaluated. Results the expression of microRNA-18a in uterine myoma was significantly lower than that in normal smooth muscle tissue adjacent to tumor (P 0.05), while the expression of ER 偽 in uterine myoma was significantly higher than that in normal smooth muscle tissue adjacent to tumor (P 0.05). There was a negative correlation between the expression of microRNA-18a and ER 偽 in uterine myoma and normal smooth muscle tissues adjacent to the tumor. Conclusion microRNA-18a may play an important role in the occurrence and development of uterine myoma by affecting the expression of ER 偽 in uterine myoma, and microRNA-18a may be a new target for the treatment of uterine myoma.
