子宮內(nèi)膜基質(zhì)干細胞的體外分離培養(yǎng)及生物學(xué)特性

【摘要】:背景:從干細胞角度去認識和研究子宮內(nèi)膜細胞,能夠為臨床上解決子宮內(nèi)膜引發(fā)的子宮內(nèi)膜疾病治療提供新的治療方案和研究切入點。目的:比較不同方法體外分離、培養(yǎng)子宮內(nèi)膜基質(zhì)干細胞的生物學(xué)特性。方法:采用3種常用的細胞分離方法(胰蛋白酶法、膠原酶Ⅰ法、胰蛋白酶聯(lián)合膠原酶Ⅰ法)從切除的子宮中分離培養(yǎng)內(nèi)膜基質(zhì)干細胞,通過組織分離培養(yǎng),對生長5 d的第2代細胞進行錐蟲藍染色,計數(shù)活細胞數(shù)量。采用間質(zhì)干細胞標志物CD146聯(lián)合造血干細胞標志物CD90單克隆抗體,以免疫組化及RT-PCR法對培養(yǎng)獲得的子宮內(nèi)膜基質(zhì)干細胞進行鑒定;同時取生長對數(shù)期細胞進行培養(yǎng),并繪制細胞生長曲線。結(jié)果與結(jié)論:(1)培養(yǎng)獲得的子宮內(nèi)膜基質(zhì)干細胞形態(tài)呈現(xiàn)梭形,細胞貼壁生長,排列無極性,具有成纖維細胞形態(tài);(2)子宮內(nèi)膜基質(zhì)干細胞標志物CD146和CD90的DNA分別在500 bp和150 bp呈現(xiàn)高表達,而子宮肌層組織對照組不表達;(3)培養(yǎng)所得干細胞的生長曲線呈現(xiàn)S型,3種方法培養(yǎng)的細胞生長周期差異無顯著性意義;相對于胰蛋白酶法和胰蛋白酶聯(lián)合膠原酶Ⅰ法而言,膠原酶Ⅰ法分離培養(yǎng)所得的子宮內(nèi)膜基質(zhì)干細胞數(shù)量較多,生長速度較快;(4)結(jié)果表明,從切除的子宮組織中能夠成功分離出子宮內(nèi)膜基質(zhì)細胞,且膠原酶Ⅰ法是一種較為高效、實用、穩(wěn)定的體外子宮內(nèi)膜基質(zhì)干細胞培養(yǎng)方法。
