蟾蜍靈對(duì)宮頸癌細(xì)胞的增殖抑制作用及其機(jī)制研究

【摘要】:背景與目的:宮頸癌仍然是婦科腫瘤致死病因的第二位,部分原因?yàn)榛熌退帯s蛤莒`是傳統(tǒng)中藥蟾酥的成分之一,目前在國(guó)內(nèi)廣泛應(yīng)用于腫瘤的治療中。該研究旨在探討蟾蜍靈對(duì)宮頸癌細(xì)胞ME180和C33A的增殖抑制作用及相關(guān)機(jī)制。方法:采用CCK8(cell counting kit-8)法檢測(cè)蟾蜍靈對(duì)宮頸癌細(xì)胞增殖的抑制作用,采用葡萄糖檢測(cè)分析試劑盒(glucose assay kit)進(jìn)行細(xì)胞內(nèi)糖代謝檢測(cè)分析,采用實(shí)時(shí)熒光定量聚合酶鏈反應(yīng)(quantitative real-time polymerase chain reaction,q RT-PCR)檢測(cè)細(xì)胞內(nèi)葡萄糖轉(zhuǎn)運(yùn)蛋白1(glucose transporter1,GLUT1)和己糖激酶2(hexokinase 2,HK2)的m RNA的表達(dá)水平,采用蛋白[質(zhì)]印跡法(Western blot)檢測(cè)原癌基因C-MYC和缺氧誘導(dǎo)因子1α(Hif-1α)表達(dá)的變化。結(jié)果:CCK8法結(jié)果顯示,蟾蜍靈能明顯抑制宮頸癌細(xì)胞ME180和C33A細(xì)胞增殖(P=0.027,P=0.018)。糖代謝檢測(cè)結(jié)果顯示,蟾蜍靈處理組葡萄糖代謝水平顯著降低(P=0.034,P=0.036)。q RT-PCR檢測(cè)結(jié)果顯示,蟾蜍靈處理組糖代謝相關(guān)指標(biāo)GLUT1(P=0.019,P=0.016)和HK2(P=0.039,P=0.041)的表達(dá)水平明顯下調(diào)。Western blot檢測(cè)結(jié)果顯示,蟾蜍靈處理后宮頸癌細(xì)胞內(nèi)C-MYC和Hif-1α蛋白表達(dá)水平顯著下調(diào)。結(jié)論:蟾蜍靈可以通過(guò)降低宮頸癌細(xì)胞ME180和C33A糖代謝水平,從而抑制細(xì)胞增殖。
[Abstract]:Background & objective: cervical cancer is still the second leading cause of death in gynecological tumors, partly due to chemotherapy resistance. Bufalin is one of the components of traditional Chinese medicine toad pastry, which is widely used in the treatment of tumor in China. The purpose of this study was to investigate the inhibitory effect of bufalin on the proliferation of cervical cancer cells ME180 and C33A and its related mechanism. Methods: CCK8 (cell counting kit-8 assay was used to detect the inhibitory effect of bufalin on the proliferation of cervical cancer cells, and glucose assay kit (glucose assay kit) was used to detect intracellular glucose metabolism. The expression of m RNA in intracellular glucose transporter 1 (glucose transporter1,GLUT1) and hexokinase 2 (hexokinase 2, HK2) was detected by real time fluorescence quantitative polymerase chain reaction (quantitative real-time polymerase chain reaction,q RT-PCR). The expression of proto-Oncogene C-MYC and hypoxia inducible factor 1 偽 (Hif-1 偽) was detected by (Western blot).
