【摘要】：目的:研究辣椒堿對(duì)Hela(HPV+)和C-33A(HPV-)細(xì)胞生長(zhǎng)增殖和凋亡的影響,探討辣椒堿對(duì)宮頸癌細(xì)胞的抑制作用與相關(guān)機(jī)制。方法:選取HPV(+)的Hela和HPV(-)的C-33A宮頸癌細(xì)胞株為研究對(duì)象。CCK-8法檢測(cè)細(xì)胞活性,流式細(xì)胞術(shù)檢測(cè)辣椒堿對(duì)細(xì)胞生長(zhǎng)的抑制作用、細(xì)胞周期和對(duì)細(xì)胞凋亡的影響;q PCR和Western blot法檢測(cè)細(xì)胞凋亡相關(guān)基因m RNA和蛋白表達(dá)。結(jié)果:辣椒堿對(duì)宮頸癌Hela和C-33A細(xì)胞增殖抑制呈濃度和時(shí)間依賴性;辣椒堿能顯著促進(jìn)兩種細(xì)胞凋亡,對(duì)Hela細(xì)胞的促凋亡效果更明顯。辣椒堿處理后,兩種細(xì)胞的細(xì)胞色素C(cytchrome C)和caspase3表達(dá)量顯著增加,Bax表達(dá)量增加,Bcl-2表達(dá)量降低。結(jié)論:辣椒堿可顯著抑制宮頸癌Hela和C-33A細(xì)胞增殖和促進(jìn)細(xì)胞的凋亡,其機(jī)理可能是通過誘導(dǎo)細(xì)胞內(nèi)活性氧類物質(zhì)(ROS)增加、細(xì)胞色素C和Caspase3表達(dá)量增加等途徑,最終誘導(dǎo)細(xì)胞凋亡。
