CIITA、LGMN、CTSB和GILT基因在子宮頸癌免疫逃逸中的作用

【摘要】:目的通過檢測抗原呈遞過程中CIITA、LGMN、CTSB、GILT基因表達模式的變化,探討抗原呈遞過程在宮頸癌免疫逃逸機制中的作用。方法收集子宮頸鱗癌、子宮頸上皮內瘤變(CIN)2和正常宮頸組織石蠟包埋標本各7例。采用QuantiGene2.0Reagent System定量檢測CⅡTA、LGMN、CTSB和GILT基因的表達,cluster3.0聚類分析其基因表達模式特征。結果 CⅡTA、GILT基因在宮頸癌和CIN組織中呈較高表達,正常宮頸中呈較低表達;LGMN基因在宮頸癌和CIN組織中呈較低表達,而在正常宮頸中表達稍高;CTSB基因表達變異較大,在異常組織中表達可上調或下降,而正常組織中表達中等。結論四個基因可顯示免疫系統(tǒng)抗原呈遞的特征,可能區(qū)分CIN和宮頸鱗癌,有助宮頸癌的預診,但由于例數(shù)少,有待進一步驗證。
