CIITA、LGMN、CTSB和GILT基因在子宮頸癌免疫逃逸中的作用

【摘要】:目的通過檢測抗原呈遞過程中CIITA、LGMN、CTSB、GILT基因表達(dá)模式的變化,探討抗原呈遞過程在宮頸癌免疫逃逸機(jī)制中的作用。方法收集子宮頸鱗癌、子宮頸上皮內(nèi)瘤變(CIN)2和正常宮頸組織石蠟包埋標(biāo)本各7例。采用QuantiGene2.0Reagent System定量檢測CⅡTA、LGMN、CTSB和GILT基因的表達(dá),cluster3.0聚類分析其基因表達(dá)模式特征。結(jié)果 CⅡTA、GILT基因在宮頸癌和CIN組織中呈較高表達(dá),正常宮頸中呈較低表達(dá);LGMN基因在宮頸癌和CIN組織中呈較低表達(dá),而在正常宮頸中表達(dá)稍高;CTSB基因表達(dá)變異較大,在異常組織中表達(dá)可上調(diào)或下降,而正常組織中表達(dá)中等。結(jié)論四個(gè)基因可顯示免疫系統(tǒng)抗原呈遞的特征,可能區(qū)分CIN和宮頸鱗癌,有助宮頸癌的預(yù)診,但由于例數(shù)少,有待進(jìn)一步驗(yàn)證。
