孕期酒精暴露抑制低氧環(huán)境下新生大鼠耗氧量和基本節(jié)律性呼吸放電

【摘要】:目的研究孕期酒精暴露對低氧環(huán)境下新生大鼠在體耗氧量和離體延髓腦片基本節(jié)律性呼吸放電(RRDA)有無抑制作用。方法成年大鼠分為酒精暴露組和對照組,酒精暴露組從合籠前3天開始以8%酒精水溶液為唯一飲用水源,對照組正常飲食,其余飼養(yǎng)條件相同,使用兩組孕鼠產(chǎn)下的新生大鼠:1采用體積描記法測量2~10 d新生大鼠在空氣、15%氧濃度、10%氧濃度、5%氧濃度下單位時(shí)間內(nèi)耗氧量,比較兩組新生大鼠在不同氧濃度下耗氧量有無差異;2使用2 d新生大鼠制作離體延髓腦片標(biāo)本,記錄RRDA,依次使用經(jīng)95%氧O2+5%二氧化碳CO2、80%O2+4.2%CO2+15.8%氮N2、65%O2+3.4%CO2+31.6%N2、50%O2+2.6%CO2+47.4%N2充分飽和的0℃改良kreb’s(MKS)從高氧到低氧灌流腦片,記錄并分析各組腦片RRDA的改變。結(jié)果 1在體耗氧量實(shí)驗(yàn)顯示在空氣、15%氧濃度、10%氧濃度下兩組新生大鼠單位時(shí)間內(nèi)耗氧量差異無統(tǒng)計(jì)學(xué)意義,在5%氧濃度下,酒精暴露組耗氧量低于對照組。2酒精暴露組腦片放電弱于對照組,即RRDA的吸氣時(shí)程(TI)、呼吸周期(RC)、放電積分幅度(IA)均弱于對照組。3隨著灌流液氧濃度下降,兩組腦片放電均逐漸減弱,TI縮短、RC延長、IA降低,酒精暴露組RRDA降低程度大于對照組。結(jié)論 1孕期酒精暴露抑制新生大鼠在低氧濃度下攝取氧的能力及離體延髓腦片RRDA。2兩組新生大鼠整體耗氧量在5%氧濃度下差異有統(tǒng)計(jì)學(xué)意義,而離體腦片放電在80%氧濃度即下降的實(shí)驗(yàn)結(jié)果間接證實(shí)外周化學(xué)感受器對呼吸有著重要的調(diào)節(jié)作用。
[Abstract]:Objective to study the inhibitory effects of alcohol exposure during pregnancy on oxygen consumption in vivo and basic rhythmic respiratory discharge (RRDA) in isolated medulla oblongata slices of neonatal rats in hypoxic environment. Methods Adult rats were divided into two groups: alcohol exposure group and control group. The alcohol exposure group was treated with 8% alcohol solution as the only drinking water source from 3 days before cages. The control group had a normal diet and the other feeding conditions were the same. Two groups of newborn rats were given birth. 1 oxygen consumption in air, 15% oxygen concentration, 10% oxygen concentration and 5% oxygen concentration in air was measured by volume drawing method. The oxygen consumption at different oxygen concentrations was compared between the two groups. (2) in vitro medulla oblongata slices were made from neonatal rats for 2 days, and RRDA, was recorded.
