妊娠期糖尿病大鼠胰腺組織中p-PERK、ATF4和CHOP蛋白的表達(dá)

【摘要】:目的:探討磷酸化蛋白激酶R樣內(nèi)質(zhì)網(wǎng)激酶(p-PERK)、C/EBP同源蛋白(CHOP)、ATF4蛋白的表達(dá)與高脂高糖誘導(dǎo)的妊娠期糖尿病大鼠胰島β細(xì)胞凋亡的關(guān)系。方法:30只雌性SD大鼠分為模型組(高脂高糖喂養(yǎng))和對(duì)照組(普食喂養(yǎng)),每組15只,妊娠第21天行靜脈糖耐量及靜脈胰島素釋放實(shí)驗(yàn);取胰腺組織行HE染色和病理組織學(xué)觀察,TUNEL法檢測(cè)胰島β細(xì)胞凋亡情況,免疫組化法檢測(cè)胰腺組織中p-PERK蛋白的表達(dá),Western blot檢測(cè)胰腺組織中ATF4及CHOP蛋白的表達(dá)。結(jié)果:模型組大鼠第一時(shí)相胰島素分泌受抑制,胰島素分泌峰值延遲;病理組織學(xué)可見模型組大鼠胰島形態(tài)欠規(guī)則,數(shù)目減少不明顯,部分細(xì)胞空泡變性,毛細(xì)血管擴(kuò)張、充血,有出血發(fā)生。模型組大鼠胰腺組織細(xì)胞凋亡率較對(duì)照組升高(t=21.164,P0.001)。與對(duì)照相比,模型組大鼠胰腺組織p-PERK、ATF4及CHOP蛋白表達(dá)水平明顯升高(t=44.178、49.249、39.664,P0.001);模型組大鼠胰腺組織中ATF4與CHOP蛋白的表達(dá)呈正相關(guān)(r=0.810,P=0.004)。結(jié)論:PERK、ATF4可能參與妊娠期糖尿病大鼠胰島β細(xì)胞凋亡的發(fā)生。
[Abstract]:Aim: to investigate the relationship between the expression of phosphorylated protein kinase R-like endoplasmic reticulum kinase (p-PERK), C/EBP homologous protein (CHOP), ATF4 protein and islet 尾 -cell apoptosis in gestational diabetic rats induced by hyperlipidemia and hyperglycemia. Methods: thirty female SD rats were divided into two groups: model group (fed with high fat and high sugar) and control group (15 rats in each group). HE staining and histopathological observation were performed in pancreatic tissues. Apoptosis of islet 尾 cells was detected by TUNEL method. The expression of p-PERK protein in pancreatic tissue was detected by immunohistochemical method. The expression of ATF4 and CHOP protein in pancreatic tissue was detected by, Western blot. Results: the first phase of insulin secretion was inhibited and the peak value of insulin secretion was delayed in the model group. Histopathology showed that the number of islets in the model group was not regular, the number of islets was not obvious, some cells were vacuolar degeneration, capillary dilatation, hyperemia and haemorrhage occurred. The apoptosis rate of pancreatic tissue in the model group was higher than that in the control group (t = 21.164, P 0.001). Compared with the control group, the expression of p-PERKN ATF4 and CHOP protein in the model group was significantly higher than that in the control group. There was a positive correlation between the expression of ATF4 and CHOP protein in the pancreatic tissue of the model group (r = 0.810 P0. 004). Conclusion: PERK,ATF4 may be involved in the apoptosis of islet 尾 cells in gestational diabetic rats.
