ACh誘導(dǎo)的臍帶血管收縮效應(yīng)及其機(jī)制研究

【摘要】:[目的]研究毒蕈堿受體激動(dòng)劑乙酰膽堿(ACh, acetylcholine)對(duì)正常人臍帶血管的舒縮功能的影響及其相應(yīng)機(jī)制并與其對(duì)大鼠和羊血管舒縮功能的作用進(jìn)行比較研究。 [方法](1)臨床收集正常足月新生兒臍帶;(2)收集孕21天胎鼠臍帶,分別分離活性離體動(dòng)靜脈臍帶血管;(3)收集正常成年雄性大鼠胸主動(dòng)脈、腸系膜動(dòng)脈,孕晚期胎羊頸動(dòng)脈,腸系膜動(dòng)脈和股動(dòng)脈。分別將分離后的血管剪成3-5mm長(zhǎng)度的血管環(huán),置于5ml Krebs37℃恒溫水浴槽內(nèi),進(jìn)行大血管和微血管功能實(shí)驗(yàn)測(cè)定ACh對(duì)血管張力的影響。同時(shí),本實(shí)驗(yàn)采用RT-PCR技術(shù)檢測(cè)了人臍血管中M受體亞型的分布。 【結(jié)果】ACh、卡巴膽堿cabarchol (10-11-10-3mol/L)對(duì)臍帶血管表現(xiàn)為較強(qiáng)的劑量依賴性收縮作用,而且在低濃度時(shí)也并未觀察到其對(duì)臍帶血管的舒張作用。ACh、cabarchol對(duì)臍帶血管的收縮作用可被阿托品(10-6mo1/L)完全阻斷,在基礎(chǔ)血管張力下,尼古丁(nicotine,10-11-10"3mol/L)對(duì)臍帶血管張力沒有明顯影響,但通過抑制血管內(nèi)皮NO合成可使臍帶血管對(duì)ACh的收縮反應(yīng)增強(qiáng),而PKC通路阻斷劑和L-Ca通道阻斷劑則可降低ACh的收縮反應(yīng)。RT-PCR結(jié)果顯示,除M1受體在臍帶血管中表達(dá)量較少以外,M2-M5的rn-RNA表達(dá)水平基本相似。 【結(jié)論】ACh對(duì)正常人臍帶血管表現(xiàn)為較強(qiáng)的收縮作用而不是舒張作用,實(shí)驗(yàn)表明ACh對(duì)臍帶血管的收縮作用與M受體下游的L-Ca、PKC通路有關(guān)。
