HCMV感染對宮頸癌細胞Siha發(fā)生上皮-間質(zhì)轉(zhuǎn)化的作用

【摘要】:目的研究人巨細胞病毒(HCMV)感染對宮頸癌細胞Siha發(fā)生上皮-間質(zhì)轉(zhuǎn)化的作用及意義。方法實驗組用HCMV AD169(MOI=5)和5ng/ml TGF-β1作用宮頸癌Siha細胞,同時將僅TGF-β1作用的Siha細胞作為對照組,分別在0、24、48、72h觀察細胞形態(tài)的變化,應用RT-PCR檢測E-鈣粘蛋白和波形蛋白的轉(zhuǎn)錄水平,采用Western blot檢測E-鈣粘蛋白、波形蛋白和MMP-2的表達水平。結(jié)果實驗組細胞于48h發(fā)生細胞形態(tài)學的變化,早于對照組的72h;實驗組上皮細胞標記物E-鈣粘蛋白的轉(zhuǎn)錄和表達水平較對照組顯著下調(diào)(P0.01),間質(zhì)細胞標記物波形蛋白的轉(zhuǎn)錄和表達較對照組顯著上調(diào)(P0.01)且表達提前,MMP-2的表達較對照組顯著上調(diào)(P0.01)且表達提前。結(jié)論 HCMV感染可促進宮頸癌細胞Siha發(fā)生化上皮-間質(zhì)轉(zhuǎn)化。
[Abstract]:Objective to study the effect and significance of human cytomegalovirus (HCMV) infection on epithelial-interstitial transformation of cervical cancer cell Siha. Methods HCMV AD169 (MOI=5) and 5ng/ml TGF- 尾 1 were used to treat cervical cancer Siha cells in the experimental group and Siha cells treated with TGF- 尾 1 alone were used as control group. The morphological changes of the cells were observed at 48 h after treatment. The transcription levels of E-cadherin and vimentin were detected by RT-PCR, and the expression of Ecadherin, vimentin and MMP-2 by Western blot. Results the morphological changes of the cells in the experimental group were observed at 48h, earlier than those in the control group (72 h). The transcription and expression level of E-cadherin in the experimental group was significantly lower than that in the control group (P0.01), and the expression of vimentin in the interstitial cells was up-regulated (P0.01), and the expression of vimentin in the interstitial cells was earlier than that in the control group (P0.01). The expression of MMP-2 was significantly up-regulated than that of control group (P0.01). Conclusion HCMV infection can promote the epithelial-interstitial transformation of Siha in cervical cancer cells.
