白假絲酵母菌最優(yōu)化RNA提取策略的探索

【摘要】:目的探索白假絲酵母菌總RNA簡單、快捷、高效的提取方法。方法選取昆明醫(yī)科大學(xué)第一附屬醫(yī)院婦產(chǎn)科門診2011年7月—2013年3月有臨床癥狀且陰道分泌物鏡檢陽性的外陰陰道假絲酵母菌病(VVC)患者的白假絲酵母菌40株,選用不同時間與溫度組合下的4種溶壁酶(Lyticase)破壁方案(A方案:37℃、12 h,B方案:25℃、12 h,C方案:37℃、24 h,D方案:25℃、24 h)進行RNA提取,將所得RNA進行熒光定量PCR(q PCR)反應(yīng)驗證RNA的濃度及純度,并進行管家基因18S rRNA的檢測。結(jié)果 D方案提取獲得的RNA濃度高于A、B、C方案(P0.05);B方案提取獲得的RNA濃度高于A、C方案(P0.05);而A、C兩方案提取獲得的RNA濃度比較,差異無統(tǒng)計學(xué)意義(P0.05)。B、D方案獲得的RNA純度高于A、C方案(P0.05);但A、C方案獲得的RNA純度比較,差異無統(tǒng)計學(xué)意義(P0.05);B方案提取獲得的RNA純度高于D方案(P0.05)。A、B、C、D方案提取的RNA擴增產(chǎn)物Ct轉(zhuǎn)化值比較,差異有統(tǒng)計學(xué)意義(P0.05);其中B、D方案q PCR擴增產(chǎn)物Ct轉(zhuǎn)化值高于A、C方案(P0.05);但B、D方案及A、C方案之間的q PCR擴增產(chǎn)物Ct轉(zhuǎn)化值比較,差異無統(tǒng)計學(xué)意義(P0.05)。管家基因18S rRNA的引物擴增出的產(chǎn)物片段大小為150 bp。結(jié)論以Lyticase破壁提取白假絲酵母菌具有簡單易行及RNA產(chǎn)量高、純度好的優(yōu)點,值得臨床推廣應(yīng)用。
