金雀異黃素對(duì)圍絕經(jīng)期小鼠卵巢和子宮組織中芳香化酶和FSHR的調(diào)控作用

【摘要】:目的:研究金雀異黃素對(duì)圍絕經(jīng)期模型小鼠卵巢和子宮組織中芳香化酶P450及促卵泡成熟激素受體(FSHR)的調(diào)節(jié)作用。方法 :選擇12月齡自然衰老ICR小鼠作為圍絕經(jīng)期動(dòng)物模型,分為對(duì)照組(CG),Gen低(L-Gen)、中(M-Gen)、高劑量組(H-Gen)及雌激素組(EG)。劑量組分別灌胃金雀異黃素(Gen)15,30,60 mg/kg,雌激素組灌胃戊酸雌二醇0.5 mg/kg,持續(xù)8周。RT-PCR及熒光定量PCR法測(cè)定小鼠卵巢和子宮中P450arom mRNA,CYP19 mRNA和FSHR的表達(dá)水平。結(jié)果 :與對(duì)照組相比,H-Gen和EG小鼠P450arom mRNA,CYP19mRNA和FSHR的表達(dá)量均升高,差異顯著(P0.05)。結(jié)論 :金雀異黃素(60 mg/kg)可使小鼠卵巢與子宮中P450arom mRNA,CYP19 mRNA和FSHR的表達(dá)量升高,雌激素作用顯著,揭示金雀異黃素可通過(guò)調(diào)節(jié)芳香化酶和促卵泡成熟激素受體基因來(lái)改善圍絕經(jīng)期卵巢與子宮的功能。
[Abstract]:Aim: to study the effects of genistein on aromatase P450 and follicle maturation hormone receptor (FSHR) in ovarian and uterine tissues of perimenopausal model mice. Methods: 12-month-old natural aging ICR mice were selected as perimenopausal animal models and divided into control group (L-Gen), moderate (M-Gen), high dose group (H-Gen) and estrogen group (EG). The estradiol valerate was perfused for 8 weeks with estradiol valerate 0.5 mg/kg, in the estrogen group. The expression of P450arom mRNA,CYP19 mRNA and FSHR in ovary and uterus of mice were measured by RT-PCR and fluorescence quantitative PCR method. Results: compared with the control group, the expression of P450arom mRNA,CYP19mRNA and FSHR in H-Gen and EG mice increased significantly (P0.05). Conclusion: genistein (60 mg/kg) can increase the expression of P450arom mRNA,CYP19 mRNA and FSHR in mouse ovary and uterus. It is suggested that genistein can improve the function of ovary and uterus by regulating aromatase and follicle stimulating hormone receptor gene.
