【摘要】：目的探討茶多酚對(duì)人卵巢癌SKOV3和3AO細(xì)胞黏附能力及對(duì)上皮型鈣黏蛋白(epithelial cadherin,E-cadherin)和神經(jīng)型鈣黏蛋白(neural cadherin,N-cadherin)表達(dá)的影響。方法體外培養(yǎng)人卵巢癌SKOV3和3AO細(xì)胞,采用MTT法檢測(cè)0、5、10、20、40、80和160μg/mL不同濃度茶多酚處理24h后,SKOV3和3AO細(xì)胞體外增殖情況,選取后續(xù)實(shí)驗(yàn)茶多酚的給藥濃度。分別選取5、10及20μg/mL茶多酚處理SKOV3細(xì)胞24h,10、20及40μg/mL茶多酚處理3AO細(xì)胞24h,各自對(duì)照組為不含茶多酚的培養(yǎng)液培養(yǎng)24h,采用細(xì)胞分離實(shí)驗(yàn)觀察細(xì)胞-細(xì)胞間黏附能力的變化;采用細(xì)胞黏附實(shí)驗(yàn)觀察細(xì)胞-基質(zhì)間黏附能力的變化;利用Real-time PCR和蛋白質(zhì)印跡法檢測(cè)E-cadherin和N-cadherin在mRNA及蛋白水平的表達(dá)變化。結(jié)果 MTT實(shí)驗(yàn)結(jié)果顯示,SKOV3(χ2=18.96,P=0.004)和3AO(χ2=16.98,P=0.009)卵巢癌細(xì)胞組內(nèi)增殖抑制率隨濃度增高而增強(qiáng)。細(xì)胞分離實(shí)驗(yàn)結(jié)果顯示,隨著茶多酚給藥濃度的增加,SKOV3和3AO細(xì)胞在10和20min的分離率逐漸降低,差異均有統(tǒng)計(jì)學(xué)意義,P值均0.05。細(xì)胞黏附率的比較,SKOV3細(xì)胞對(duì)照組、5、10和20μg/mL組的黏附率分別為100.00%、93.89%、82.50%和67.32%,各組間差異有統(tǒng)計(jì)學(xué)意義,χ2=9.583,P=0.022;3AO細(xì)胞對(duì)照組、10、20和40μg/mL組黏附率分別為100.00%、92.65%、86.90%和76.71%,差異有統(tǒng)計(jì)學(xué)意義,χ2=10.532,P=0.015。Real-time PCR結(jié)果顯示,SKOV3細(xì)胞各實(shí)驗(yàn)組的E-cadherin mRNA表達(dá)水平增加,χ2=10.532,P=0.015,N-cadherin mRNA表達(dá)水平減少,χ2=9.583,P=0.022;3AO細(xì)胞中各組N-cadherin mRNA表達(dá)水平明顯減少,χ2=10.532,P=0.015,但E-cadherin mRNA表達(dá)水平無(wú)明顯變化,χ2=4.499,P=0.212。蛋白質(zhì)印跡法結(jié)果顯示,SKOV3細(xì)胞中各實(shí)驗(yàn)組的E-cadherin蛋白表達(dá)水平增加,χ2=10.116,P=0.018,N-cadherin蛋白表達(dá)水平減少,χ2=9.804,P=0.02;3AO細(xì)胞中實(shí)驗(yàn)組與對(duì)照比較N-cadherin蛋白表達(dá)水平明顯減少,χ2=8.868,P=0.031,但E-cadherin蛋白表達(dá)水平差異無(wú)統(tǒng)計(jì)學(xué)意義,χ2=1.17,P=0.76。結(jié)論茶多酚能夠提高卵巢癌SKOV3和3AO細(xì)胞-細(xì)胞間黏附能力,降低細(xì)胞-基質(zhì)間黏附能力,其機(jī)制可能與茶多酚上調(diào)E-cadherin及下調(diào)N-cadherin表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effects of tea polyphenols on the adhesion of human ovarian cancer SKOV3 and 3AO cells and on the expression of epithelial cadherin (epithelial cadherin,E-cadherin) and neuronal cadherin (neural cadherin,N-cadherin). Methods Human ovarian cancer SKOV3 and 3AO cells were cultured in vitro. The proliferation of SKOV3 and 3AO cells was detected by MTT assay after different concentrations of tea polyphenols were treated with different concentrations of tea polyphenols for 24 hours. The concentration of tea polyphenols in subsequent experiments was selected. 10 and 20 渭 g/mL tea polyphenols were used to treat SKOV3 cells for 24 h and 40 渭 g/mL tea polyphenols for 24 h, respectively. The control groups were cultured in culture medium without tea polyphenols for 24 h. Cell separation experiments were used to observe the change of cell-cell adhesion ability. Cell adhesion assay was used to observe the change of cell-matrix adhesion ability, and the expression of E-cadherin and N-cadherin in mRNA and protein levels were detected by Real-time PCR and Western blotting. Results the results of MTT assay showed that the inhibitory rate of proliferation in SKOV3 (蠂 2 + 18.96) and 3AO (蠂 2 + 16.98) ovarian cancer cells increased with the increase of the concentration. The results of cell separation experiment showed that with the increase of tea polyphenol concentration, the isolation rate of SKOV3 and 3AO cells at 10 and 20min decreased gradually, and the difference was statistically significant (P < 0.05). The adhesion rate of SKOV3 cells in the control group, 5 渭 g/mL group and 20 渭 g/mL group was 100.0098.89% and 67.322.50%, respectively. There was significant difference between the two groups (蠂 2, 9.583, P0. 022, P < 0. 022, P 0. 022, P 0. 022). The adhesion rates of 3AO cells in the control group, 100.20 渭 g/mL group and 40 渭 g/mL group were 86.90% and 76.71%, respectively. The difference was statistically significant. The results of 蠂 ~ 2 + 10.532P ~ (0.015) Real-time PCR showed that there was no significant difference between the two groups. The expression level of E-cadherin mRNA was increased in each experimental group of SKOV3 cells, and the expression level of N-cadherin mRNA was decreased in 蠂 2 + 10.532 (P = 0.015), 蠂 ~ (2 +) 9.583 (P ~ (0.022); The expression level of N-cadherin mRNA was significantly decreased in 3AO cells, 蠂 2 + 10.532% P0. 015, but E-cadherin mRNA expression level was not significantly changed. The results of Western blotting showed that the expression level of E-cadherin protein was increased in each experimental group of SKOV3 cells, and the expression level of 蠂 ~ 2 ~ (10. 116) ~ (10. 116) Pu 0.018 ~ (N-cadherin) protein decreased, and that of 蠂 ~ (2) 9.804 ~ (?) Compared with the control group, the expression of N-cadherin protein in the experimental group was significantly lower than that in the control group, and the expression level of E-cadherin protein in the experimental group was significantly lower than that in the control group (蠂 2 + 8.868). However, there was no significant difference in the expression level of E-cadherin protein between the experimental group and the control group. Conclusion Tea polyphenols can increase the ability of SKOV3 and 3AO cell-cell adhesion and decrease the ability of cell-matrix adhesion. The mechanism may be related to the up-regulation of E-cadherin and the down-regulation of N-cadherin expression by tea polyphenols.
