【摘要】：子宮內(nèi)膜損傷是導(dǎo)致宮腔粘連的最主要原因,建立有效的子宮內(nèi)膜損傷動(dòng)物模型是研究此類疾病發(fā)生、發(fā)展和治療反應(yīng)等不可或缺的支撐條件。通過宮腔注射95%的乙醇制造大鼠(Rattus norvegicus)子宮內(nèi)膜損傷模型,檢測胚胎植入數(shù)目來分析子宮內(nèi)膜損傷對(duì)大鼠生育情況的影響,觀察大鼠子宮內(nèi)膜厚度、腺體數(shù)量和纖維化面積,分析乙醇處理對(duì)子宮內(nèi)膜的影響;檢測細(xì)胞角蛋白(CK-19)和波性蛋白(Vimentin)的表達(dá)水平,分析上皮細(xì)胞和間質(zhì)細(xì)胞的增殖分化及子宮內(nèi)膜損傷程度。結(jié)果顯示,正常組大鼠子宮比損傷組更為平滑且有韌性,與正常組相比,損傷組大鼠生育率顯著降低(P0.01),子宮內(nèi)膜厚度變薄(P0.01)、腺體數(shù)量顯著減少(P0.01),纖維化面積顯著增大(P0.01),CK-19和Vimentin表達(dá)量顯著下調(diào)。結(jié)果提示已成功建立大鼠子宮內(nèi)膜損傷動(dòng)物模型。
[Abstract]:Endometrial injury is the main cause of uterine cavity adhesion. Establishing an effective animal model of endometrial injury is an indispensable supporting condition to study the occurrence, development and therapeutic response of this kind of disease. The (Rattus norvegicus) endometrial injury model of rats was made by intrauterine injection of 95% ethanol. The number of embryo implantation was measured to analyze the effect of endometrial injury on the fertility of rats. The thickness of endometrium, the number of glands and the area of fibrosis were observed. The effects of ethanol treatment on endometrium were analyzed, and the expression levels of cytokeratin (CK-19) and wave protein (Vimentin) were detected, and the proliferation and differentiation of epithelial cells and interstitial cells were analyzed as well as the degree of endometrial injury. The results showed that the uterus of the normal group was smoother and more resilient than that of the injured group. In the injured group, the fertility rate decreased significantly (P0.01), the thickness of endometrium became thinner (P0.01), the number of glands decreased significantly (P0.01), the area of fibrosis increased significantly (P0.01), and the expression of CK-19 and Vimentin decreased significantly. The results suggest that the rat model of endometrial injury has been successfully established.
