HIF-1α、CXCR4及VEGF在子宮內(nèi)膜癌組織中的表達(dá)及意義

【摘要】:目的探究缺氧誘導(dǎo)因子(HIF)-1α及其下游基因趨化因子受體(CXCR)4、血管內(nèi)皮生長因子(VEGF)在子宮內(nèi)膜癌組織與正常子宮內(nèi)膜組織中的表達(dá)及其意義。方法用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)及免疫組織化學(xué)技術(shù)檢測94例子宮內(nèi)膜癌組織和30例正常子宮內(nèi)膜組織中HIF-1α、CXCR4、VEGF mRNA及蛋白的表達(dá)情況,并分析其與子宮內(nèi)膜癌臨床病理特征的關(guān)系。結(jié)果 HIF-1α、CXCR4、VEGF mRNA及蛋白在子宮內(nèi)膜癌組織中存在高表達(dá),而在正常子宮內(nèi)膜組織中幾乎無表達(dá)。HIF-1α、CXCR4蛋白表達(dá)在正常子宮內(nèi)膜組織中的表達(dá)與子宮內(nèi)膜癌組織比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。HIF-1α和CXCR4蛋白的表達(dá)水平與子宮內(nèi)膜癌的脈管浸潤、臨床分期及患者術(shù)后生存時間相關(guān)。結(jié)論 HIF-1α、CXCR4及VEGF在子宮內(nèi)膜癌組織中表達(dá)水平增高,提示HIF-1α可能通過調(diào)控CXCR4及VEGF蛋白的轉(zhuǎn)錄,進(jìn)而促進(jìn)腫瘤組織新生血管生成而促進(jìn)子宮內(nèi)膜癌的發(fā)展。
[Abstract]:Objective to investigate the expression and significance of hypoxia inducible factor (HIF) 1 偽 and its downstream chemokine receptor (CXCR) 4 and vascular endothelial growth factor (VEGF) in endometrial carcinoma and normal endometrium. Methods the expression of HIF-1 偽, CXCR4,VEGF mRNA and protein in 94 cases of endometrial carcinoma and 30 cases of normal endometrium were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. Results HIF-1 偽, CXCR4,VEGF mRNA and protein were highly expressed in endometrial carcinoma. The expression of HIF-1 偽 and CXCR4 protein in normal endometrial tissue was significantly different from that in endometrial carcinoma tissue (P0.05). The expression levels of HIF-1 偽 and CXCR4 protein were correlated with the vascular infiltration of endometrial carcinoma. Clinical stage and postoperative survival time were correlated. Conclusion the expression of HIF-1 偽, CXCR4 and VEGF in endometrial carcinoma is increased, which suggests that HIF-1 偽 may promote the development of endometrial carcinoma by regulating the transcription of CXCR4 and VEGF proteins and thus promoting angiogenesis.
