【摘要】：目的:觀察穿心蓮內(nèi)酯對卵巢癌細胞株SKOV-3侵襲與凋亡的影響并探討初步的作用機制。方法:CCK-8法檢測不同濃度(0、5、10、20和40μmol/L)的穿心蓮內(nèi)酯對SKOV-3細胞作用不同時間(12、24、36和48h)后,SKOV-3細胞存活率的變化;Transwell法與TUNEL法分別檢測SKOV-3細胞侵襲能力與凋亡能力的變化;Western blot法檢測p-PI3K、p-Akt與p-mTOR蛋白水平的變化。結果:CCK-8法檢測結果顯示,隨著濃度增加與培養(yǎng)時間延長,穿心蓮內(nèi)酯對SKOV-3細胞存活率的抑制程度增強;采用20μmol/L穿心蓮內(nèi)酯培養(yǎng)SKOV-3細胞36h后,SKOV-3細胞的侵襲數(shù)明顯降低,凋亡數(shù)明顯增加(P0.05),p-PI3K、p-Akt與p-mTOR的蛋白水平明顯降低(P0.05)。結論:穿心蓮內(nèi)酯能夠抑制卵巢癌細胞SKOV-3的活力與侵襲能力,增強凋亡能力,這可能與抑制PI3K/Akt/mTOR信號通路有關。
[Abstract]:Aim: to investigate the effect of andrographolide on invasion and apoptosis of ovarian cancer cell line SKOV-3. Methods the changes of survival rate of SKOV-3 cells after different concentrations of andrographolide (0 5, 10, 10, 20 and 40 渭 mol/L) at different time points (12: 24, 36 and 48 hours) were detected by 10% CCK-8 method. Transwell and TUNEL methods were used to detect the invasion and apoptosis of SKOV-3 cells respectively. Western blot assay was used to detect the changes of cell invasion and apoptosis of SKOV-3 cells. The changes of p-PI3 KT p-Akt and p-mTOR protein levels were measured. Results the inhibition of andrographolide on the survival rate of SKOV-3 cells was increased with the increase of concentration and culture time, and the invasion number of SKOV-3 cells was decreased after 36 h culture with 20 渭 mol/L andrographolide. The number of apoptosis was significantly increased (P0.05), the protein levels of p-PI3KT p-Akt and p-mTOR were significantly decreased (P0.05). Conclusion: andrographolide can inhibit the activity, invasion and apoptosis of SKOV-3 cells, which may be related to the inhibition of PI3K/Akt/mTOR signaling pathway.
