【摘要】：目的:探討Dicer1在正常宮頸上皮、宮頸上皮內(nèi)瘤變和宮頸鱗癌的表達(dá)及意義。方法:Western blot和RT-PCR法檢測正常細(xì)胞系(End1/E6E7)、宮頸內(nèi)瘤變細(xì)胞系(S12)及宮頸癌細(xì)胞系(Ca Ski、He La、Si Ha和C33A)中Dicer1蛋白及其mRNA表達(dá)水平,免疫組織化學(xué)技術(shù)檢測Dicer1在正常宮頸、宮頸上皮內(nèi)瘤變和宮頸鱗癌中的表達(dá)水平。結(jié)果:正常宮頸上皮細(xì)胞系Dicer1表達(dá)水平低于宮頸癌細(xì)胞系表達(dá)量。隨著惡性程度增加,Dicer1呈駝峰狀表達(dá),即在正常宮頸組織及Grade III級宮頸鱗癌中表達(dá)較低,但在宮頸上皮內(nèi)瘤變及高分化宮頸腫瘤(grade I級)中表達(dá)較高。結(jié)論:Dicer1在正常宮頸、宮頸上皮內(nèi)瘤變和宮頸鱗癌中的表達(dá)有差異,為進(jìn)一步研究Dicer1在宮頸癌發(fā)生發(fā)展中的作用及分子機(jī)制提供依據(jù)。
[Abstract]:Objective: to investigate the expression and significance of Dicer1 in normal cervical epithelium, cervical intraepithelial neoplasia and cervical squamous cell carcinoma. Methods the expression of Dicer1 protein and mRNA in normal cell line (End1/E6E7), cervical carcinoma cell line (S12) and cervical carcinoma cell line (Ca Ski,He La,Si Ha and C33A) were detected by Western blot and RT-PCR. The expression of Dicer1 in normal cervix was detected by immunohistochemistry. Expression level of cervical intraepithelial neoplasia and cervical squamous cell carcinoma. Results: the expression level of Dicer1 in normal cervical epithelial cell line was lower than that in cervical carcinoma cell line. The expression of Dicer1 in normal cervix and Grade III grade cervical squamous cell carcinoma was lower with the increase of malignant degree, but was higher in (grade I grade of cervical intraepithelial neoplasia and highly differentiated cervical carcinoma. Conclusion there are differences in the expression of Dicer1 in normal cervix, cervical intraepithelial neoplasia and cervical squamous cell carcinoma, which provides a basis for further study of the role and molecular mechanism of Dicer1 in the carcinogenesis and development of cervical cancer.
